卵巢漿液性癌組織中microRNA表達(dá)譜分析

【摘要】:目的尋找與卵巢漿液性癌相關(guān)的microRNA,積累以國內(nèi)患者為數(shù)據(jù)來源的microRNA表達(dá)譜資料,并初步探討其臨床意義。方法利用microRNA芯片,篩查卵巢漿液性癌組織及正常卵巢組織中差異表達(dá)的microRNA,用實(shí)時熒光定量PCR在58份卵巢組織標(biāo)本中進(jìn)行驗(yàn)證,并分析microRNA表達(dá)水平與卵巢漿液性癌臨床病理特征之間的關(guān)系。結(jié)果與正常卵巢組織比較,芯片結(jié)果顯示卵巢漿液性癌有9個microRNA明顯上調(diào),分別是hsa-mi R-141-3p、hsa-mi R-200a-3p、hsa-mi R-200b-3p、hsa-mi R-200c-3p、hsa-mi R-224-5p、hsa-mi R-7-5p、hsa-mi R-142-3p、hsa-mi R-21-5p、hsa-mi R-142-5p,6個microRNA明顯下調(diào),分別是hsa-mi R-424-5p、hsa-mi R-214-3p、hsa-mi R-125b-5p、hsa-let-7b-5p、hsa-mi R-199a-5p、hsa-mi R-100-5p。實(shí)時熒光定量PCR驗(yàn)證結(jié)果與芯片結(jié)果一致。對差異microRNA進(jìn)行分層分析發(fā)現(xiàn),與卵巢漿液性癌Ⅰ-Ⅱ期、淋巴結(jié)未轉(zhuǎn)移患者比較,Ⅲ-Ⅳ期、淋巴結(jié)有轉(zhuǎn)移患者的hsa-mi R-21-5p表達(dá)均增高,hsa-mi R-125b-5p表達(dá)均降低(P0.05);而在高級別與低級別卵巢癌患者之間microRNA的表達(dá)水平差異均無統(tǒng)計(jì)學(xué)意義(P均0.05)。結(jié)論卵巢漿液性癌中異常表達(dá)的microRNA可能與腫瘤的發(fā)生發(fā)展相關(guān)。
