【摘要】：目的探討乳腺癌新輔助化療(neoadjuvant chemotherapy,NCT)前后自噬的變化及其與XIAP變化的關(guān)系。方法應(yīng)用免疫組化En Vision兩步法檢測(cè)NCT前后94對(duì)乳腺癌組織中p62和XIAP的表達(dá),觀察NCT后p62和XIAP表達(dá)的變化情況,分析其相關(guān)性。結(jié)果 NCT前乳腺癌標(biāo)本中,p62高表達(dá)率為51.1%(48/94),XIAP高表達(dá)率為84.0%(79/94);NCT后乳腺癌標(biāo)本中,p62高表達(dá)率升高為81.9%(77/94),XIAP高表達(dá)率降低為35.1%(33/94)。即NCT后p62高表達(dá)率明顯升高,XIAP高表達(dá)率明顯降低(P均0.05)。NCT前后相比,p62表達(dá)升高的樣本率為73.4%,XIAP表達(dá)下降的樣本率為75.5%,且p62表達(dá)變化與XIAP表達(dá)變化呈顯著負(fù)相關(guān)(P0.01)。結(jié)論乳腺癌NCT后自噬增強(qiáng)與XIAP下調(diào)共存,且XIAP可能參與NCT后自噬增強(qiáng)的負(fù)性調(diào)節(jié)。
[Abstract]:Objective to investigate the changes of autophagy in breast cancer before and after neoadjuvant chemotherapy (neoadjuvant) and the relationship between autophagy and XIAP. Methods the expression of p62 and XIAP in 94 cases of breast cancer before and after NCT was detected by immunohistochemical en Vision two-step method. The expression of p62 and XIAP was observed after NCT, and the correlation between p62 and XIAP was analyzed. Results the high expression rate of p62 in breast cancer specimens before NCT was 51.1% (48 / 94). The high expression rate of XIAP was 84.0% (79 / 94). The high expression rate of p62 was increased to 81.9% (77 / 94) and decreased to 35.1% (33 / 94). That is to say, the high expression rate of p62 was significantly increased after NCT. The high expression rate of XIAP decreased significantly (P0. 05). The sample rate of increased p62 expression before and after NCT was 73.5%, and the change of p62 expression was negatively correlated with the change of XIAP expression (P0.01). Conclusion autophagy after NCT in breast cancer coexists with down-regulation of XIAP, and XIAP may participate in the negative regulation of autophagy after NCT.
