HSPA2在胰腺癌中的表達及其臨床意義

【摘要】:目的檢測并對比熱休克蛋白A2(HSPA2)在胰腺癌及癌旁胰腺組織中的表達水平,分析HSPA2蛋白在胰腺癌標(biāo)本中的表達水平與其臨床病理參數(shù)之間的相關(guān)性。方法采用實時定量PCR、Western blot法檢測并對比胰腺癌及癌旁胰腺組織中HSPA2蛋白和mRNA的表達水平,采用免疫組織化學(xué)染色方法測定胰腺癌患者組織中HSPA2蛋白的表達,并探討其表達與臨床病理特征之間的關(guān)系。結(jié)果與癌旁胰腺組織相比,胰腺癌組織中HSPA2 mRNA和蛋白表達水平均明顯升高。HSPA2蛋白在胰腺癌組織中的陽性表達率為68.8%(55/80)。HSPA2蛋白表達量與胰腺癌的分化程度、血管侵犯、TNM分期差異有統(tǒng)計學(xué)意義(P=0.011、0.005、0.012),而與患者性別、年齡、腫塊大小及位置、血清CA199水平差異無統(tǒng)計學(xué)意義。結(jié)論 HSPA2在胰腺癌組織中的表達明顯高于與之相應(yīng)的癌旁胰腺組織,且HSPA2的高表達與胰腺癌的惡性進展、侵襲和轉(zhuǎn)移等事件密切相關(guān)。
[Abstract]:Objective to detect the expression level of specific heat shock protein A2 (HSPA2) in pancreatic cancer and adjacent pancreatic tissue, and to analyze the correlation between the expression level of HSPA2 protein in pancreatic cancer specimens and its clinicopathological parameters. Methods the real-time quantitative PCR, Western blot method were used to detect and compare the HSPA2 protein and M in pancreatic cancer and the pancreatic tissue adjacent to the cancer. The expression level of RNA was measured by immunohistochemical staining, and the expression of HSPA2 protein in the tissues of pancreatic cancer patients was measured and the relationship between the expression and the clinicopathological features was discussed. The results showed that the expression level of HSPA2 mRNA and protein in pancreatic cancer tissues significantly increased the positive of.HSPA2 protein in pancreatic cancer tissues. The expression rate of 68.8% (55/80).HSPA2 protein was significantly different from that of pancreatic cancer, vascular invasion and TNM staging (P=0.011,0.005,0.012), but there was no significant difference between the sex, age, the size and position of the tumor and the level of CA199 in the patients. Conclusion the expression of HSPA2 in the pancreatic cancer tissues is significantly higher than that of the same. The high expression of HSPA2 in adjacent pancreatic tissues is closely related to the malignant progression, invasion and metastasis of pancreatic cancer.
