CXCL12與Wnt2b蛋白在膽管癌組織中的中的表達(dá)及其臨床意義

【摘要】:目的:本研究通過(guò)免疫組織化學(xué)法檢測(cè)膽管癌組織及非惡性膽管系統(tǒng)疾病組織中CXCL12與Wnt2b蛋白的表達(dá),并探討CXCL12與Wnt2b蛋白在膽管癌組織中的表達(dá)情況與性別、年齡、組織分化程度、膽管癌TNM分期及是否有淋巴結(jié)轉(zhuǎn)移之間的關(guān)系。方法:實(shí)驗(yàn)采用以抗原、抗體相結(jié)合為原理的免疫組織化學(xué)染色法,對(duì)CXCL12、Wnt2b在膽管癌及非惡性膽道系統(tǒng)疾病組織中的表達(dá)進(jìn)行測(cè)定,并分析其與膽管癌相關(guān)病理特征之間的關(guān)系。入選及排除標(biāo)準(zhǔn):排除其他部位惡性腫瘤病史者,且術(shù)前未接受放化療治療。統(tǒng)計(jì)學(xué)方法:應(yīng)用χ2檢驗(yàn)檢測(cè)兩種蛋白的陽(yáng)性表達(dá)率是否存在差異,并分析數(shù)據(jù)相關(guān)性,所有數(shù)據(jù)應(yīng)用統(tǒng)計(jì)學(xué)軟件進(jìn)行統(tǒng)計(jì)學(xué)分析,當(dāng)P0.05時(shí),認(rèn)為差異具有統(tǒng)計(jì)學(xué)意義。結(jié)果:(1).在膽管癌組織及非惡性膽管系統(tǒng)疾病組織中CXCL12蛋白的陽(yáng)性表達(dá)具有顯著差異(P0.05),分別為:89.2%、23.3%;(2).在膽管癌組織及非惡性膽管系統(tǒng)疾病組織中Wnt2b蛋白的陽(yáng)性表達(dá)同樣具有顯著差異(P0.05),分別為:86.2%、16.7%;(3).CXCL12在膽管癌組織中的表達(dá)與患者性別、年齡、組織分化程度、淋巴結(jié)轉(zhuǎn)移、TNM分期無(wú)關(guān)(P0.05);(4).Wnt2b在膽管癌組織中的表達(dá)與患者性別、年齡無(wú)關(guān)(P0.05),與組織分化程度、淋巴結(jié)轉(zhuǎn)移、TNM分期有關(guān)聯(lián)(P0.05),即在組織分化程度為中低分化、淋巴結(jié)轉(zhuǎn)移組、TNM為Ⅲ、Ⅳ期組呈高表達(dá);(5).CXCL12與Wnt2b的表達(dá)呈正相關(guān)(r=0.28,P0.05)。結(jié)論:(1).CXCL12與Wnt2b兩種蛋白可能均參與了膽管癌疾病的發(fā)生、發(fā)展、浸潤(rùn)與轉(zhuǎn)移過(guò)程;(2).兩種蛋白聯(lián)合檢測(cè)的方法可以更加準(zhǔn)確的進(jìn)行疾病的診斷、組織分型、TNM分期及淋巴結(jié)轉(zhuǎn)移的判定。
[Abstract]:Objective: to study the expression of CXCL12 and Wnt2b protein in cholangiocarcinoma and non-malignant bile duct diseases by immunohistochemical method, and to investigate the expression of CXCL12 and Wnt2b in cholangiocarcinoma, sex and age. Relationship between tissue differentiation, TNM staging of cholangiocarcinoma and lymph node metastasis. Methods: the expression of CXCL12Wnt2b in cholangiocarcinoma and non-malignant biliary tract diseases was determined by immunohistochemical staining based on antigen and antibody, and the relationship between CXCL12Wnt2b and the pathological features of cholangiocarcinoma was analyzed. Inclusion and exclusion criteria: patients with history of malignant tumors in other parts were excluded and were not treated with radiotherapy and chemotherapy before operation. Statistical method: 蠂 2 test was used to detect the difference of positive expression rate of the two proteins, and the data correlation was analyzed. All data were analyzed by statistical software. When P0.05, the difference was considered statistically significant. Results: (1). The positive expression of CXCL12 protein was significantly different in cholangiocarcinoma tissues and non-malignant cholangiocarcinoma tissues (P0.05), which were respectively: (1) 89. 2% and 23. 3%; (2). The positive expression of Wnt2b protein was also significantly different in cholangiocarcinoma tissues and non-malignant cholangiocarcinoma tissues (P0.05), respectively, and the expression of CXCL12 in cholangiocarcinoma tissues was significantly higher than that in patients with cholangiocarcinoma (P 0.05), and the expression of CXCL12 in cholangiocarcinoma tissues was correlated with the sex, age and degree of tissue differentiation.
