【摘要】：目的:探討胱天蛋白酶募集域蛋白9(caspase recruitment domain containing protein 9,CARD9)在乳腺癌中的表達及其臨床意義。方法:應用實時熒光定量PCR和蛋白質印跡法檢測乳腺癌細胞、正常乳腺細胞、乳腺癌組織和癌旁組織中CARD9 mRNA和蛋白的表達,分析CARD9表達與乳腺癌患者臨床病理特征間的關系。應用實時熒光定量PCR法檢測乳腺癌組織和癌旁組織中核因子κB(nuclear factor-κB,NF-κB)信號通路相關因子p50、p65、白細胞介素1β(interlukin-1β,IL-1β)和IL-12 mRNA的表達,蛋白質印跡法檢測乳腺癌組織和癌旁組織中p50、p65、抑制性NF-κB激酶α(inhibitor of NF-κB kinaseα,IKKα)和磷酸化IKKα(phosphorylated-IKKα,p-IKKα)蛋白的表達。WST-1法檢測CARD9高表達對乳腺癌細胞增殖的影響。結果:乳腺癌組織中CARD9 mRNA(P0.01)和蛋白(P=0.012)表達水平顯著高于癌旁組織。乳腺癌T-47D細胞(P=0.021)和MCF-7細胞(P=0.014)中CARD9 mRNA表達水平高于正常乳腺細胞Hs578Bst,乳腺癌ZR-75-1細胞中CARD9 mRNA(P=0.003)和蛋白(P=0.015)表達水平均高于正常乳腺細胞Hs578Bst。CARD9蛋白表達與腫瘤大小和雌激素受體(estrogen receptor,ER)表達有關(P值均0.05)。乳腺癌組織中p50(P0.05)和p-IKKα(P0.01)蛋白的表達水平高于癌旁組織,而且乳腺癌組織中CARD9 mRNA表達與p50(r=0.622,P0.001)、p65(r=0.392,P=0.005)、IL-1β(r=0.685,P0.001)和IL-12(r=0.556,P0.001)mRNA表達間呈正相關。CARD9高表達可促進乳腺癌細胞的增殖(P0.05)。結論:CARD9可能通過激活NF-κB信號通路而促進細胞增殖,參與乳腺癌的發(fā)生和發(fā)展。
[Abstract]:Objective: to investigate the expression and clinical significance of cystatin recruitment domain protein 9 (caspase recruitment domain containing protein 9 CARD9 in breast cancer. Methods: the expression of CARD9 mRNA and protein in breast cancer cells, normal breast cells, breast cancer tissues and adjacent tissues were detected by real-time fluorescence quantitative PCR and Western blot. The relationship between CARD9 expression and clinicopathological features of breast cancer patients was analyzed. The expression of nuclear factor- 魏 B (NF- 魏 B) signaling transduction factors p50 p65, interleukin-1 尾 (interlukin-1 尾 -IL-1 尾) and IL-12 mRNA in breast cancer and adjacent tissues were detected by real-time fluorescence quantitative PCR assay. The effects of high expression of CARD9 on the proliferation of breast cancer cells were detected by Western blot. The expression of p50 p65, inhibitory NF- 魏 B kinase 偽 (inhibitor of NF- 魏 B kinase 偽 Ike 偽 and phosphorylated IKK 偽 (phosphorylated-IKK 偽 -pIKK 偽) in breast cancer tissues and adjacent tissues were detected by WST-1 method. Results: the expression levels of CARD9 mRNA (P0.01) and protein (P0. 012) in breast cancer tissues were significantly higher than those in paracancerous tissues. The expression of CARD9 mRNA in breast cancer T-47D cells (Pn0.021) and MCF-7 cells (Pn0.014) was higher than that in normal breast cells Hs578Bst.Breast cancer ZR-75-1 cells expressed CARD9 mRNA (Pn0.003) and protein (Pn0.015) higher than those in normal breast cells, tumor size and estrogen receptor. The expression of estrogen receptor ER was related (P < 0. 05). The expression levels of p50 (P0.05) and p-IKK 偽 (P0.01) in breast cancer tissues were higher than those in adjacent tissues, and the expression of CARD9 mRNA was positively correlated with the expression of p50 (0.392p0.001) p65 (0.392P ~ (0.005) and IL-12 (r 0.556P ~ (0.001) mRNA (P0.05). Conclusion the cell proliferation may be promoted by the activation of NF- 魏 B signaling pathway, and may be involved in the occurrence and development of breast cancer.
