卡鉑短期誘導人卵巢癌類干細胞的分離及初步鑒定

【摘要】:目的分離卡鉑耐藥型人卵巢癌類干細胞,為研發(fā)和篩選針對卡鉑耐藥的卵巢癌二線治療藥物提供細胞模型。方法從卵巢癌組織或腹水中分離培養(yǎng)卵巢癌細胞,直接或采用卡鉑短期誘導后無血清懸浮培養(yǎng),分離純化獲得卵巢癌類干細胞球,采用免疫熒光法或流式細胞術(shù)檢測干細胞標志物的表達。結(jié)果卵巢癌原代細胞直接或采用卡鉑短期誘導后無血清懸浮培養(yǎng)能夠形成類干細胞球,并高表達干細胞標記物CD133,ALDH1,ABCG2和CD24?ㄣK短期誘導分離的卵巢癌干細胞球中CD24+細胞含量明顯增加。結(jié)論原代培養(yǎng)卵巢癌細胞經(jīng)卡鉑短期誘導后無血清懸浮培養(yǎng),能進一步富集純化卵巢癌類干細胞,可作為卵巢癌耐藥研究的理想細胞模型。
[Abstract]:Objective to isolate carboplatin resistant human ovarian cancer stem cells and to provide a cell model for the development and screening of carboplatin resistant second line therapy drugs for ovarian cancer. Methods Ovarian cancer cells were isolated and cultured from ovarian cancer tissues or ascites. The expression of stem cell markers was detected by immunofluorescence or flow cytometry. Results Serum-free suspension culture of primary ovarian cancer cells, either directly or by carboplatin for a short time, could result in the formation of stem cell like spheres and overexpression of stem cell markers CD133, ALDH1, ABCG2 and CD24. The content of CD24 cells in ovarian cancer stem cell spheres induced by carboplatin was significantly increased. Conclusion Serum-free suspension culture of primary cultured ovarian cancer cells induced by carboplatin for a short time can further enrich and purify ovarian cancer like stem cells and can be used as an ideal cell model for the study of ovarian cancer drug resistance.
