HER2抑制上皮鈣黏素表達(dá)促進(jìn)人乳腺上皮細(xì)胞遷移

【摘要】:目的研究原癌基因人表皮生長(zhǎng)因子受體2(HER2)對(duì)MCF10A人乳腺上皮細(xì)胞形態(tài)和遷移能力的影響及機(jī)制。方法通過慢病毒包裝及感染MCF10A人乳腺上皮細(xì)胞,建立HER2過表達(dá)細(xì)胞,用TranswellTM法研究細(xì)胞遷移能力變化,結(jié)合Western blot法、實(shí)時(shí)定量PCR和免疫熒光染色檢測(cè)細(xì)胞中上皮鈣黏素(E-cadherin)的表達(dá)。結(jié)果過表達(dá)HER2的乳腺上皮細(xì)胞呈分散型生長(zhǎng),細(xì)胞遷移能力增強(qiáng),E-cadherin蛋白表達(dá)受到抑制,細(xì)胞膜上E-cadherin表達(dá)也顯著降低,重新過表達(dá)E-cadherin能抑制HER2誘導(dǎo)的細(xì)胞遷移和分散。結(jié)論 HER2能通過抑制E-cadherin促進(jìn)人乳腺上皮細(xì)胞遷移。
[Abstract]:Objective to investigate the effect and mechanism of proto-oncogene human epidermal growth factor receptor 2 (HER2) on the morphology and migration of MCF10A human mammary epithelial cells. Methods HER2 overexpression cells were established by packaging and infecting MCF10A human mammary epithelial cells by lentivirus. Transwell TM method was used to study the changes of cell migration ability. The expression of E-cadherin was detected by real-time quantitative PCR and immunofluorescence staining with Western blot. Results the overexpression of HER2 in breast epithelial cells was characterized by dispersive growth. The expression of E-cadherin protein was inhibited and the expression of E-cadherin was decreased significantly. E-cadherin reexpression could inhibit the migration and dispersion induced by HER2. Conclusion Her2 can promote the migration of human mammary epithelial cells by inhibiting E-cadherin.
