【摘要】：目的:研究Runx2對(duì)肝癌細(xì)胞中MMP9的表達(dá)以及其對(duì)肝癌細(xì)胞侵襲遷移能力的影響。方法:通過(guò)免疫組織化學(xué)染色法對(duì)2005年12月至2015年12月天津醫(yī)科大學(xué)腫瘤醫(yī)院和天津醫(yī)科大學(xué)總醫(yī)院189例肝細(xì)胞肝癌病例標(biāo)本中Runx2和MMP9的表達(dá)進(jìn)行分析;將Runx2過(guò)表達(dá)質(zhì)粒和干擾質(zhì)粒分別轉(zhuǎn)染至肝癌細(xì)胞系HepG2和SMMC7721細(xì)胞中。使用Western blot法檢測(cè)轉(zhuǎn)染后HepG2和SMMC7721細(xì)胞中Runx2和MMP9的表達(dá)情況;劃痕、侵襲實(shí)驗(yàn)檢測(cè)Runx2對(duì)肝癌細(xì)胞遷移和侵襲能力的影響。結(jié)果:免疫組織化學(xué)結(jié)果顯示Runx2的表達(dá)和MMP9的表達(dá)相關(guān)。在HepG2細(xì)胞中上調(diào)Runx2的表達(dá),促進(jìn)了MMP9的表達(dá),增強(qiáng)了HepG2細(xì)胞遷移侵襲能力;在SMMC7721細(xì)胞中下調(diào)Runx2的表達(dá),降低了MMP9的表達(dá),抑制了SMMC7721細(xì)胞的遷移侵襲能力。結(jié)論:Runx2的表達(dá)可能參與促進(jìn)肝細(xì)胞肝癌細(xì)胞MMP9的表達(dá),進(jìn)而促進(jìn)肝細(xì)胞肝癌的遷移侵襲能力。
[Abstract]:Aim: to study the effect of Runx2 on the expression of MMP9 and the ability of invasion and migration of hepatoma cells. Methods: from December 2005 to December 2015, the expression of Runx2 and MMP9 in 189 cases of hepatocellular carcinoma (HCC) in Tianjin Medical University Oncology Hospital and Tianjin Medical University General Hospital were analyzed by immunohistochemical staining. Runx2 overexpression plasmid and interference plasmid were transfected into hepatoma cell lines HepG2 and SMMC7721, respectively. The expression of Runx2 and MMP9 in transfected HepG2 and SMMC7721 cells was detected by Western blot assay, and the effect of Runx2 on the migration and invasion of hepatoma cells was detected by scratch and invasion assay. Results: immunohistochemical results showed that the expression of Runx 2 was correlated with the expression of MMP9. The expression of Runx2 was up-regulated in HepG2 cells, which promoted the expression of MMP9 and enhanced the ability of migration and invasion of HepG2 cells, and down-regulated the expression of Runx2 in SMMC7721 cells, reduced the expression of MMP9 and inhibited the migration and invasion of SMMC7721 cells. Conclusion the expression of 1: Runx2 may play an important role in promoting the expression of MMP9 in hepatocellular carcinoma cells, thus promoting the ability of migration and invasion of hepatocellular carcinoma.
