【摘要】：目的探討早期乳腺浸潤性導(dǎo)管癌中microRNA-10b(mi R-10b)表達(dá)與臨床指標(biāo)及預(yù)后關(guān)系。方法采用原位分子雜交技術(shù),檢測305例中早期乳腺浸潤性導(dǎo)管癌組織miR-10b表達(dá),采用Pearson χ~2檢驗(yàn)分析miR-10b表達(dá)在不同臨床指標(biāo)間的差異性;采用Spearman秩和相關(guān)分析早期乳腺浸潤性導(dǎo)管癌中miR-10b、年齡、淋巴結(jié)、雌激素受體α(ERα)、孕激素受體(PR)及人類表皮生長因子受體2(HER-2)之間的相關(guān)性。采用Kaplan-Meier法計(jì)算累計(jì)無瘤生存時間,應(yīng)用Log-rank檢驗(yàn)比較miR-10b不同表達(dá)組間無瘤生存時間的差異。結(jié)果早期乳腺浸潤性導(dǎo)管癌中miR-10b表達(dá)與年齡、腫瘤大小和復(fù)發(fā)轉(zhuǎn)移,差異具有統(tǒng)計(jì)學(xué)意義(P0.05);與月經(jīng)狀態(tài)、臨床分期、化療、放療及內(nèi)分泌治療比較,差異無統(tǒng)計(jì)學(xué)意義(P0.05)。miR-10b陽性表達(dá)與HER-2陽性表達(dá)呈正相關(guān),差異有統(tǒng)計(jì)學(xué)意義(P0.05)。miR-10b表達(dá)陰性中位無瘤生存時間較miR-10b表達(dá)陽性時間長,兩者比較差異有統(tǒng)計(jì)學(xué)意義(P0.05);淋巴結(jié)陰性時miR-10b陰性表達(dá)的中位無瘤生存時間較陽性表達(dá)長,兩者比較差異有統(tǒng)計(jì)學(xué)意義(P0.05);相反,miR-10b表達(dá)在淋巴結(jié)陽性中位無瘤生存時間無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 miR-10b表達(dá)水平與早期乳腺浸潤性導(dǎo)管癌復(fù)發(fā)轉(zhuǎn)移相關(guān),特別在無淋巴結(jié)轉(zhuǎn)移且miR-10b陽性表達(dá)組的中位無瘤生存時間短,預(yù)后差。miR-10b陽性表達(dá)是早期乳腺浸潤性導(dǎo)管癌的預(yù)后因素。
[Abstract]:Objective to investigate the relationship between the expression of microRNA-10bmmi R-10b in early invasive ductal carcinoma of breast and the clinical indexes and prognosis. Methods the expression of miR-10b in 305 cases of breast invasive ductal carcinoma was detected by in situ hybridization. The difference of miR-10b expression among different clinical indexes was analyzed by Pearson 蠂 2 test. The correlation between miR-10b, age, lymph node, estrogen receptor 偽 -ER 偽, progesterone receptor (PRP) and human epidermal growth factor receptor (2HER-2) in early invasive ductal carcinoma of breast was analyzed by Spearman rank sum correlation. Kaplan-Meier method was used to calculate the cumulative tumor-free survival time, and Log-rank test was used to compare the difference of tumor-free survival time among different miR-10b expression groups. Results the expression of miR-10b was significantly different from age, tumor size, recurrence and metastasis in early breast invasive ductal carcinoma (P 0.05), and compared with menstrual status, clinical stage, chemotherapy, radiotherapy and endocrine therapy. There was no significant difference between the positive expression of P0.05, miR-10b and the positive expression of HER-2. There was a significant difference between the positive expression of HER-2 and the positive expression of HER-2. The median tumor-free survival time of the negative expression of P0.05, miR-10b was longer than that of the positive expression of miR-10b. The median tumor-free survival time of miR-10b negative expression in lymph node negative was longer than that of positive expression. The difference between the two groups was statistically significant (P 0.05), whereas the expression of miR-10b was not significant in lymph node positive median tumor-free survival time (P 0.05). Conclusion the expression level of miR-10b is associated with the recurrence and metastasis of early invasive ductal carcinoma of the breast, especially in the group with no lymph node metastasis and miR-10b positive expression. Poor prognosis. MiR-10b positive expression is the prognostic factor of early invasive ductal carcinoma of breast.
