【摘要】：目的早期食管癌臨床癥狀常不明顯,腫瘤標(biāo)志物的深入研究對(duì)食管癌早期診斷極為重要。文中通過研究體外特定細(xì)胞株[Eca-109及人食管正常上皮細(xì)胞(HEEC)]中Rock2和Wnt11蛋白表達(dá)量,分析各蛋白的表達(dá)與食管癌的關(guān)系。方法培養(yǎng)Eca-109和HEEC細(xì)胞株,RT-q PCR和Western blot技術(shù)檢測(cè)Rock2和Wnt11在細(xì)胞株中的表達(dá)水平。結(jié)果RT-q PCR檢測(cè)結(jié)果顯示,與HEEC相比,Eca-109中Rock2 mRNA、Wnt11 mRNA表達(dá)水平明顯升高[(1.000±0.000)vs(4.955±0.539),(1.000±0.000)vs(2.925±0.230),P0.01]。Western blot檢測(cè)顯示Rock2和Wnt11蛋白在Eca-109中的表達(dá)較HEEC均升高[(955.000±21.628)vs(778.844±102.193),(2 175.316±145.623)vs(1 312.233±50.734),P0.05]。結(jié)論 Rock2和Wnt11表達(dá)增高可能是食管鱗狀細(xì)胞癌轉(zhuǎn)移的標(biāo)志。
[Abstract]:Objective the clinical symptoms of early esophageal carcinoma are not obvious, and the further study of tumor markers is very important for early diagnosis of esophageal carcinoma. In this paper, the expression of Rock2 and Wnt11 proteins in specific cell lines [Eca-109 and human normal esophageal epithelial cells] in vitro was studied, and the relationship between the expression of these proteins and esophageal carcinoma was analyzed. Methods the expression levels of Rock2 and Wnt11 in Eca-109 and HEEC cell lines were detected by RT-q PCR and Western blot techniques. Results compared with HEEC, the expression of Rock2 mRNA-Wnt11 mRNA in Eca-109 was significantly increased [1.000 鹵0.000)vs(4.955 鹵0.539 P0.01]. Western blot analysis showed that the expression of Rock2 and Wnt11 protein in Eca-109 was significantly higher than that in HEEC [955.000 鹵21.628)vs(778.844 鹵102.1931] 2 175.316 鹵145.623)vs(1 312.233 鹵50.734 (P0.05). Conclusion the increased expression of Rock2 and Wnt11 may be a marker for metastasis of esophageal squamous cell carcinoma.
