【摘要】：目的:探討腦源性神經(jīng)營(yíng)養(yǎng)因子-反義長(zhǎng)鏈非編碼RNA(brain-derived neurotrophic factor-antisense long non-coding RNA,BDNF-AS)對(duì)人肝癌細(xì)胞增殖和侵襲能力的影響。方法:將攜帶BDNF-AS基因的重組慢病毒分別感染肝癌細(xì)胞系HepG2和97-H后,采用RT-PCR法檢測(cè)肝癌細(xì)胞中BDNF-AS和BDNF mRNA的表達(dá)水平變化。分別采用EdU摻入實(shí)驗(yàn)、AnnexinⅤ-FITC/碘化丙啶(propidium iodide,PI)雙染和Transwell小室法檢測(cè)BDNF-AS過表達(dá)對(duì)HepG2和97-H細(xì)胞增殖、凋亡和侵襲能力的影響。結(jié)果:感染攜帶BDNF-AS基因的重組慢病毒后,肝癌HepG2和97-H細(xì)胞中BDNF-AS的表達(dá)水平均明顯升高(P值均0.000 1),而BDNF mRNA的表達(dá)水平明顯降低(P0.000 1,P=0.001)。BDNF-AS過表達(dá)后,HepG2和97-H細(xì)胞的凋亡率無明顯變化(P值均0.05),但細(xì)胞增殖率明顯降低(P0.05,P=0.001),侵襲細(xì)胞數(shù)明顯減少(P值均0.000 1)。結(jié)論:BDNF-AS過表達(dá)可能對(duì)肝癌細(xì)胞的生長(zhǎng)和侵襲具有重要的負(fù)調(diào)控作用。
[Abstract]:Aim: to investigate the effects of brain-derived neurotrophic factor-antisense long chain noncoding RNA(brain-derived neurotrophic factor-antisense long non-coding RNAs BDNF-ASon on proliferation and invasion of human hepatoma cells.
