【摘要】:目的研究PM2.5暴露對人肺癌細胞A549的自噬影響探討自噬與凋亡的關(guān)聯(lián)效應(yīng)。方法 100μg/mL PM2.5分別處理A549細胞0、2、4、12、24 h,采用免疫熒光觀察處理PM2.5 24 h對細胞自噬的影響,免疫印跡法檢測PM2.5對肺癌細胞自噬和凋亡重要基因的表達的影響,運用自噬抑制劑3-甲基腺嘌呤(3-MA)處理細胞進一步明確自噬通路對細胞凋亡的作用。結(jié)果細胞自噬標志物LC3-Ⅱ蛋白表達均明顯著增強,LC3-Ⅱ/LC3-I比值升高,而凋亡蛋白Bax逐漸下調(diào)(均P0.05);免疫熒光顯示,PM2.5組細胞的綠色熒光增強;但運用細胞自噬抑制劑(3-MA)會抑制LC3Ⅱ/LC3I的轉(zhuǎn)化,促進Bax的表達。結(jié)論PM2.5可誘導肺癌細胞保護性自噬的發(fā)生,抑制自噬能夠促進PM2.5對肺癌細胞的凋亡誘導。
[Abstract]:Objective to investigate the effects of PM2.5 exposure on autophagy in human lung cancer cell line A549 and to explore the correlation between autophagy and apoptosis. Methods A549 cells were treated with 100 渭 g/mL PM2.5 for 24 h, and the effects of PM2.5 treatment on autophagy were observed by immunofluorescence. The effects of PM2.5 on the expression of autophagy and apoptosis in lung cancer cells were detected by Western blot.
