【摘要】：目的研究PM2.5暴露對(duì)人肺癌細(xì)胞A549的自噬影響探討自噬與凋亡的關(guān)聯(lián)效應(yīng)。方法 100μg/mL PM2.5分別處理A549細(xì)胞0、2、4、12、24 h,采用免疫熒光觀察處理PM2.5 24 h對(duì)細(xì)胞自噬的影響,免疫印跡法檢測(cè)PM2.5對(duì)肺癌細(xì)胞自噬和凋亡重要基因的表達(dá)的影響,運(yùn)用自噬抑制劑3-甲基腺嘌呤(3-MA)處理細(xì)胞進(jìn)一步明確自噬通路對(duì)細(xì)胞凋亡的作用。結(jié)果細(xì)胞自噬標(biāo)志物L(fēng)C3-Ⅱ蛋白表達(dá)均明顯著增強(qiáng),LC3-Ⅱ/LC3-I比值升高,而凋亡蛋白Bax逐漸下調(diào)(均P0.05);免疫熒光顯示,PM2.5組細(xì)胞的綠色熒光增強(qiáng);但運(yùn)用細(xì)胞自噬抑制劑(3-MA)會(huì)抑制LC3Ⅱ/LC3I的轉(zhuǎn)化,促進(jìn)Bax的表達(dá)。結(jié)論P(yáng)M2.5可誘導(dǎo)肺癌細(xì)胞保護(hù)性自噬的發(fā)生,抑制自噬能夠促進(jìn)PM2.5對(duì)肺癌細(xì)胞的凋亡誘導(dǎo)。
[Abstract]:Objective to investigate the effects of PM2.5 exposure on autophagy in human lung cancer cell line A549 and to explore the correlation between autophagy and apoptosis. Methods A549 cells were treated with 100 渭 g/mL PM2.5 for 24 h, and the effects of PM2.5 treatment on autophagy were observed by immunofluorescence.
