【摘要】：目的探討Wnt分泌蛋白(WISP)1在肺癌中的表達(dá)量及作用。方法生物信息學(xué)分析WISP1在肺癌及癌旁組織中的表達(dá)量,qRTPCR檢測(cè)肺癌組織及癌旁組織中WISP1 mRNA的表達(dá)。在肺癌細(xì)胞中干預(yù)WISP1的表達(dá),檢測(cè)處理后細(xì)胞化療敏感性的變化。結(jié)果生物信息學(xué)分析免疫組化數(shù)據(jù)庫(kù)聯(lián)合qRT-PCR結(jié)果發(fā)現(xiàn),WISP1 mRNA在肺癌中的表達(dá)量顯著高于癌旁組織,且WISP1在肺癌化療耐受組的表達(dá)量顯著高于化療敏感組。在肺癌細(xì)胞中敲低WISP1對(duì)化療藥物奧沙利鉑的敏感性增加,反之亦然。結(jié)論 WISP1在肺癌中高表達(dá)且促進(jìn)肺癌化療耐受。
[Abstract]:Objective to investigate the expression and role of Wnt secretory protein (WISP) 1 in lung cancer. Methods the expression of WISP1 in lung cancer and paracancerous tissues was analyzed by bioinformatics. The expression of WISP1 mRNA in lung cancer and paracancerous tissues was detected by qRT PCR. The expression of WISP1 was interfered in lung cancer cells and the changes of chemosensitivity after treatment were detected. Results the expression of WISP1 mRNA in lung cancer was significantly higher than that in paracancerous tissues, and the expression of WISP1 in chemoresistance group was significantly higher than that in chemosensitive group. The sensitivity of WISP1 to oxaliplatin is increased in lung cancer cells and vice versa. Conclusion WISP1 is highly expressed in lung cancer and promotes chemotherapy tolerance in lung cancer.
