【摘要】：[目的]探討結(jié)直腸癌組織中信號(hào)素4D (Semaphorin4D,Sema4D)的表達(dá)及其與微血管密度(Microvessel density, MVD)的相關(guān)性。[方法]回顧性分析2014-2015年間在云南省腫瘤醫(yī)院結(jié)直腸外科接受結(jié)直腸癌根治性手術(shù)的61例患者的臨床病理資料;男性38例,女性23例,中位年齡55歲;采用免疫組織化學(xué)SP法檢測(cè)Semma4D蛋白在61例結(jié)直腸癌組織和56例正常腸粘膜組織(距癌組織邊緣5cm以上)中的表達(dá)水平;用免疫組化SP法檢測(cè)CD34標(biāo)記的微血管并計(jì)數(shù)微血管密度。[結(jié)果]Sema4D蛋白主要在結(jié)直腸癌組織的細(xì)胞膜及細(xì)胞質(zhì)內(nèi)表達(dá),在正常腸粘膜組織細(xì)胞質(zhì)內(nèi)呈陰性表達(dá)。Sema4D在癌組織的陽性表達(dá)率[100%(61/61)]明顯高于正常腸粘膜組織[0(0/56)],差異有統(tǒng)計(jì)學(xué)意義(X 2=113.027, P0.001)。結(jié)直腸癌組織中Sema4D與MVD兩者作相關(guān)性分析,Spearman相關(guān)系數(shù)r=0.333, p=0.009,兩者呈現(xiàn)明顯的正相關(guān)性;MVD表達(dá)量在Sema4D表達(dá)的高、低組之間作t檢驗(yàn)分析,p=0.003 (p0.01),差異具有顯著的統(tǒng)計(jì)學(xué)意義。[結(jié)論]Sema4D在結(jié)直腸癌組織內(nèi)高水平表達(dá),且與MVD存在明顯的正相關(guān)性,MVD隨著Sema4D表達(dá)的升高而增加,提示Sema4D有望成為潛在的結(jié)直腸癌血管治療靶點(diǎn)。
[Abstract]:[objective] to investigate the expression of semaphorin 4 D in colorectal carcinoma and its correlation with microvessel density (MVD). [methods] the clinicopathological data of 61 patients undergoing radical colorectal cancer surgery in the Department of Colorectal surgery in Yunnan Oncology Hospital from 2014 to 2015 were retrospectively analyzed, including 38 males and 23 females with median age of 55 years. Immunohistochemical SP method was used to detect the expression of Semma4D protein in 61 cases of colorectal carcinoma and 56 cases of normal intestinal mucosa (more than 5cm from the edge of cancer tissue), the microvessel labeled by CD34 was detected by SP immunohistochemical method and the microvessel density was counted. [results] Sema4D protein was mainly expressed in the cell membrane and cytoplasm of colorectal cancer. The positive rate of negative expression of Sema4D in the cytoplasm of normal intestinal mucosa was significantly higher than that in normal intestinal mucosa [100% 61 / 61] and normal intestinal mucosa [0 / 56]. The difference was statistically significant (P 0.001). Correlation Analysis of Sema4D and MVD in Colorectal Cancer; Spearman correlation coefficient r = 0.333, p = 0.009. There was a significant positive correlation between Sema4D and MVD in the expression of Sema4D, and a significant difference was found between the two groups by t test. [conclusion] there is a high level of Sema4D expression in colorectal cancer tissues, and there is a significant positive correlation between MVD and MVD, which suggests that Sema4D may be a potential target for vascular therapy for colorectal cancer.
