HDAC1、HDAC5和E-cadherin在肺腺癌中的表達(dá)和相關(guān)性研究

【摘要】:【目的】采用免疫組織化學(xué)的方法,在肺腺癌癌組織和癌旁組織中,分別對(duì)HDAC1、HDAC5和E-cadherin三種蛋白的表達(dá)水平進(jìn)行檢測(cè),同時(shí)對(duì)HDAC1、HDAC5和E-cadherin在肺腺癌癌組織和癌旁組織中的表達(dá)情況進(jìn)行對(duì)比分析,探討三種蛋白在肺腺癌組織中的表達(dá)情況與各臨床病理特征之間的關(guān)系以及三者之間共同表達(dá)的情況,為肺腺癌臨床個(gè)體化治療及尋找新的靶點(diǎn)提供相關(guān)理論依據(jù)!痉椒ā繎(yīng)用免疫組化SP法檢測(cè)在93例肺腺癌組織和48例癌旁正常肺組織中應(yīng)用免疫組織化學(xué)SP法分別檢測(cè)HDAC1、HDAC5及E-cadherin的表達(dá)結(jié)果。采用χ2檢驗(yàn)和四格表精確概率法分析HDAC1、HDAC5和E-cadherin的表達(dá)情況以及三種蛋白與肺腺癌中的變大情況與各臨床病理特征之間的關(guān)系。采用Spearman相關(guān)性分析對(duì)三種蛋白之間的相關(guān)性進(jìn)行統(tǒng)計(jì)學(xué)分析,所有實(shí)驗(yàn)所得數(shù)據(jù)均在SPSS 21.0統(tǒng)計(jì)軟件上進(jìn)行處理。P0.05則表明差異有統(tǒng)計(jì)學(xué)意義。【結(jié)果】在93例肺腺癌患者的組織中,HDAC1、HDAC5和E-cadherin的陽性表達(dá)率分別為58.06%(54/93),48.39%(45/93),45.16%(42/93),在48例癌旁組織中,HDAC1、HDAC5和E-cadherin的陽性表達(dá)率分別為20.83%(10/48),66.67%(32/48),68.75%(33/48),三種蛋白在肺腺癌組織與癌旁組織中的陽性表達(dá)率有顯著性差異(P0.05)。E-cadherin的表達(dá)與年齡及細(xì)胞分化程度密切相關(guān)(P0.05),與其他病理特征無明顯相關(guān),HDAC1的表達(dá)與年齡密切相關(guān)(P0.05),與其他病理特征無明顯相關(guān),HDAC5與臨床病理特征無明顯相關(guān)(P0.05)。HDAC1與HDAC5、HDAC1與E-cadherin之間呈負(fù)相關(guān)(r分別為-0.224、-0.236,P0.05),HDAC5與E-cadherin之間無明顯相關(guān)性(r=0.159,,P=0.128)!窘Y(jié)論】肺腺癌的發(fā)生發(fā)展是多因素、多步驟、多環(huán)節(jié)共同參與作用的復(fù)雜結(jié)果, HDAC1與HDAC5、HDAC1與E-cadherin之間呈負(fù)相關(guān),HDAC5與E-cadherin之間無明顯相關(guān)性。HDAC1的表達(dá)與年齡密切相關(guān),E-cadherin的表達(dá)與年齡及細(xì)胞分化程度密切相關(guān),聯(lián)合檢測(cè)三者可能對(duì)判斷預(yù)后、合理選擇藥物有一定指導(dǎo)意義。
[Abstract]:[objective] to detect the expression of HDAC1HDAC5 and E-cadherin in lung adenocarcinoma tissues and adjacent tissues by immunohistochemical method. At the same time, the expression of HDAC5 and E-cadherin in lung adenocarcinoma tissues and adjacent tissues were compared and analyzed, and the relationship between the expression of HDAC5 and E-cadherin in lung adenocarcinoma tissues and the clinicopathological features and the co-expression of the three proteins were discussed. [methods] Immunohistochemical SP method was used to detect 93 cases of lung adenocarcinoma and 48 cases of adjacent normal lung tissues. The expression of HDAC5 and E-cadherin were detected. The expression of HDAC5 and E-cadherin was analyzed by 蠂 2 test and quad table exact probability method. The relationship between the expression of HDAC5 and HDAC5 and the relationship between the three proteins and the enlargement of lung adenocarcinoma and the clinicopathological features were analyzed. The correlation between the three proteins was statistically analyzed by Spearman correlation analysis. All experimental data were processed on SPSS 21.0 statistical software. P05 showed significant difference. [results] in 93 cases of lung adenocarcinoma, the positive rates of HDAC5 and E-cadherin were 58.06% 58.06% 48.39% 48.39% 48.39% 43.16% 42.93% and 48 cases of paracancerous tissues were 42.93%, 58.06%, 48.39% and 48.39%, respectively, and in 48 cases of adjacent tissues, the positive rates of HDAC5 and E-cadherin were 58.06%, 48.39% and 48.39%, respectively. The positive expression rates of HDAC5 and E-cadherin were 20.83 / 10 / 48 / 66.67 / 32 / 48 and 68.7575 / 48, respectively. There was a significant difference between the expression of three proteins in lung adenocarcinoma and adjacent tissues. The expression of E-cadherin was closely related to age and cell differentiation (P0.05), but not to other pathological features. There was no significant correlation between HDAC5 expression and clinicopathological features (P 0.05). HDAC1 was negatively correlated with E-cadherin (r = -0.224- 0.236P0.05HDAC5 and E-cadherin). [conclusion] there was no significant correlation between HDAC5 and E-cadherin. [conclusion] there is no significant correlation between HDAC5 and HDAC5 and E-cadherin. [conclusion] The occurrence and development of lung adenocarcinoma are many factors. There was no significant correlation between HDAC1 and HDAC5 and E-cadherin. There was no significant correlation between HDAC5 and E-cadherin. The expression of E-cadherin was closely related to age and the degree of cell differentiation. Combined detection of the three may have certain guiding significance in judging prognosis and selecting drugs reasonably.
