晶狀體蛋白αB對卵巢癌細(xì)胞惡性行為的影響

【摘要】:目的研究晶狀體蛋白αB(αB-crystallin)在卵巢癌組織中的表達(dá)及其對卵巢癌細(xì)胞SKOV3增殖及遷移侵襲能力的影響。方法免疫組織化學(xué)法檢測人正常卵巢組織和上皮性卵巢癌組織中αB-crystallin表達(dá);針對蛋白αB-crystallin的基因(CRYAB基因)設(shè)計(jì)合成靶向小干擾RNA(small interfering RNA,si RNA)重組慢病毒,并感染人卵巢漿液性乳頭狀囊腺癌細(xì)胞株SKOV3;流式細(xì)胞儀、平板克隆、MTT檢測細(xì)胞凋亡和增殖;Transwell檢測細(xì)胞遷移和侵襲能力;Western blot檢測基質(zhì)金屬蛋白酶-2(MMP-2)和基質(zhì)金屬蛋白酶-9(MMP-9)的表達(dá)。結(jié)果αB-crystallin在卵巢癌組織中表達(dá)高于正常卵巢組織,在Ⅱ型卵巢癌組織中呈明顯高表達(dá)(P0.01),穩(wěn)轉(zhuǎn)CRYAB si RNA慢病毒后SKOV3細(xì)胞內(nèi)αB-crystallin蛋白表達(dá)降低64%,其凋亡和增殖無明顯改變,但SKOV3細(xì)胞遷移和侵襲能力明顯受到抑制(P0.01),同時(shí)蛋白MMP-2和MMP-9表達(dá)下調(diào)(P0.01)。結(jié)論αB-crystallin能夠影響卵巢癌SKOV3細(xì)胞浸潤轉(zhuǎn)移,其在Ⅱ型卵巢癌組織中的高表達(dá)可能與其高侵襲潛能相關(guān)。
[Abstract]:Objective to study the expression of 偽 B (偽 B-crystallin) in ovarian carcinoma and its effect on the proliferation and migration and invasion of ovarian cancer cell line SKOV3. Methods Immunohistochemical method was used to detect the expression of 偽 B-crystallin in human normal ovarian tissues and epithelial ovarian carcinoma tissues, and the recombinant lentivirus was designed and synthesized by targeting small interfering RNA(small interfering siRNAs targeting protein 偽 B-crystallin. The cell line SKOV3 was infected with human ovarian serous papillary cystadenocarcinoma, and the expression of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) were detected by flow cytometry and plate clone assay respectively, and the expression of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) were detected by blot. Results the expression of 偽 B-crystallin in ovarian carcinoma was higher than that in normal ovarian tissue, and the expression of 偽 B-crystallin was significantly higher in type 鈪,
