膀胱癌中EYA4基因啟動子區(qū)甲基化狀態(tài)及其臨床意義的研究

【摘要】:目的:探討膀胱癌細胞系和膀胱移行細胞癌組織中EYA4(Eyes Absent 4:EYA轉(zhuǎn)錄共激活磷酸酶4)基因啟動子區(qū)CpG島甲基化狀態(tài)及在臨床病理特征中的意義。方法:采取甲基化特異性聚合酶鏈反應(yīng)(MSP)檢測EYA4基因在5株膀胱癌細胞系、1株膀胱永生化上皮細胞系和75例膀胱移行細胞癌組織及癌旁組織中甲基化狀態(tài),并結(jié)合臨床病理特征行統(tǒng)計學分析。結(jié)果:EYA4基因啟動子區(qū)在5株膀胱癌細胞系中,有4株出現(xiàn)甲基化,其甲基化率為80%;在人正常膀胱細胞系中甲基化陰性。EYA4在膀胱癌組織中的甲基化率為70.7%(53/75),癌旁組織中為24%(18/75),差異較為顯著,有統(tǒng)計學意義(c2=32.760,P0.01)。EYA4基因啟動子區(qū)CpG島甲基化在臨床病理特征中如單發(fā)膀胱腫瘤與多發(fā)膀胱腫瘤相比,直徑≤3cm的膀胱癌腫瘤與(29)3cm的膀胱癌腫瘤相比,低級別膀胱腫瘤與高級別膀胱腫瘤相比,淺表性膀胱腫瘤與浸潤性膀胱腫瘤相比,差異均有統(tǒng)計學意義(P0.05),但是EYA4基因啟動子區(qū)CpG島甲基化程度在與膀胱癌患者的年齡和性別之間差異不明顯,無統(tǒng)計學意義(P0.05)。結(jié)論:EYA4基因啟動子區(qū)CpG島在膀胱癌細胞系和膀胱癌組織中均表現(xiàn)為高度甲基化,推測其可能與膀胱癌的發(fā)生和發(fā)展有一定的關(guān)系,將有助于膀胱癌的早期診斷。EYA4基因啟動子區(qū)CpG島區(qū)甲基化率與膀胱癌患者的臨床病理資料如腫瘤的數(shù)目、大小、分級和分期密切相關(guān),與其年齡和性別無相關(guān)性。
[Abstract]:Objective: to investigate the methylation status of CpG island in the promoter region of EYA4(Eyes Absent 4:EYA transcription co-activated phosphatase 4 gene in bladder cancer cell line and transitional cell carcinoma (TCC) and its significance in clinicopathological features. Methods: methylation-specific polymerase chain reaction (EYA4) was used to detect the methylation status of EYA4 gene in 5 bladder cancer cell lines, 1 bladder immortalized epithelial cell line and 75 bladder transitional cell carcinoma tissues and adjacent tissues. Combined with clinicopathological features, statistical analysis was made. Results methylation was found in 4 of 5 bladder cancer cell lines. Its methylation rate was 80%; in human normal bladder cell line, methylation negative. EYA4 methylation rate was 70.7% 53% 75% in bladder cancer tissue, and 24% 75% in adjacent tissue, the difference was quite significant. The methylation of CpG island in the promoter region of P0.01.
