人結(jié)直腸癌組織中TLR-4與PGE-2的表達(dá)及意義

【摘要】:目的:研究分析Toll樣受體-4(toll-like receptor 4,TLR-4)和前列腺素E2(prostaglandin E2,PGE-2)在結(jié)直腸癌組織中的表達(dá)情況,探討TLR-4和PGE-2與結(jié)直腸癌之間的相關(guān)性,對(duì)TLR-4相關(guān)信號(hào)通路的深入研究,有助于我們更好的了解消化道惡性腫瘤的發(fā)生機(jī)制,TLR-4信號(hào)通路有望成為防治消化道腫瘤的新靶點(diǎn)。方法:應(yīng)用免疫組織化學(xué)方法(S-P)法檢測(cè)40例廣西中醫(yī)藥大學(xué)患者結(jié)直腸癌、癌旁組織和遠(yuǎn)癌組織中TLR-4、PGE-2的表達(dá)。采用SPSS17.0統(tǒng)計(jì)學(xué)軟件分析處理,采用χ2檢驗(yàn),采用Spearman等級(jí)相關(guān)分析法分析TLR-4與PGE-2表達(dá)的相關(guān)性,均以P0.05為差異有統(tǒng)計(jì)學(xué)意義。結(jié)果:數(shù)據(jù)顯示TLR-4在遠(yuǎn)癌結(jié)直腸組織上呈微弱表達(dá),在癌旁組織和癌組織中呈陽性表達(dá),且在癌腫瘤中表達(dá)明顯增強(qiáng),呈散在游離或顆粒狀分布于細(xì)胞膜和細(xì)胞質(zhì)上,其定位以胞漿胞膜為主,細(xì)胞核上無表達(dá)。PGE-2在遠(yuǎn)癌結(jié)直腸組織中呈微弱表達(dá),在癌旁組織及癌組織中的表達(dá)明顯逐漸增強(qiáng),其定位也以胞漿胞膜為主,細(xì)胞核上亦無表達(dá)。TLR-4在癌組織、癌旁組織及遠(yuǎn)癌組織中的陽性面積率依次下降,分別為9.94%,5.85%,1.30%,三者相比較差異有統(tǒng)計(jì)學(xué)意義(P0.01)。PGE-2在結(jié)直腸癌組織、癌旁和遠(yuǎn)癌組織中,其表達(dá)水平逐漸下調(diào),陽性率分別為3.89%,3.19%,1.01%三者相比較差異有統(tǒng)計(jì)學(xué)意義(P0.01)。TLR-4在PGE-2陽性組中的表達(dá)率為(20/32)62.50%,PGE-2表達(dá)率為(4/8)50.00%在陰性組中;PGE-2在TLR-4陽性組中的表達(dá)率為(20/24)83.33%,在TLR-4陰性組中的表達(dá)率為(12/16)75.00%;在40例結(jié)直腸癌組織中,有20例兩者同為陽性表達(dá),4例同為陰性表達(dá),表達(dá)一致率為60.00%,分析指出兩者表達(dá)呈正相關(guān),差異有統(tǒng)計(jì)學(xué)意義(r=0.296,P0.05)。結(jié)論:1、TLR-4在結(jié)直腸癌組織、癌旁組織和遠(yuǎn)癌正常組織中的表達(dá)水平逐漸下降,距離癌組織中心越遠(yuǎn),其表達(dá)陽性面積率越低;2、PGE-2在結(jié)直腸癌組織、癌旁組織和遠(yuǎn)癌正常組織中的表達(dá)水平逐漸下降,距離癌組織中心越遠(yuǎn),其表達(dá)陽性面積率越低;3、結(jié)直腸癌組織中TLR-4和PGE-2的表達(dá)呈正相關(guān),TLR-4可能通過上調(diào)PGE-2的表達(dá)促進(jìn)結(jié)直腸癌的惡性進(jìn)展。
[Abstract]:Objective: to investigate the expression of Toll like receptor -4toll-like receptor 4 (TLR-4) and prostaglandin E2(prostaglandin E2P PGE-2 in colorectal cancer, to explore the relationship between TLR-4 and PGE-2 and colorectal cancer, and to investigate the TLR-4 related signal pathway. TLR-4 signaling pathway may be a new target for the prevention and treatment of gastrointestinal cancer. Methods: immunohistochemical method was used to detect the expression of TLR-4 and PGE-2 in 40 patients with colorectal cancer, paracancerous tissues and distant carcinomas in Guangxi University of traditional Chinese Medicine. The correlation between TLR-4 and PGE-2 expression was analyzed by SPSS17.0 statistical software, 蠂 2 test and Spearman rank correlation analysis. Results: the data showed that the expression of TLR-4 was weakly expressed in distal colorectal tissues, positive in paracancerous tissues and cancerous tissues, and increased in cancer tumors, and distributed in free or granular form on cell membrane and cytoplasm. The localization of PGE-2 was mainly in the cytoplasmic membrane, and the expression of PGE-2 was weak in the distal colorectal tissues, and increased gradually in the adjacent tissues and cancer tissues. The localization of PGE-2 was mainly in the cytosolic membrane. The positive area of TLR-4 in cancer tissues, paracancerous tissues and distal carcinomas decreased in turn, 9.945.85 and 1.30. the difference was statistically significant in colorectal cancer tissues, paracancerous tissues and distal cancer tissues, and the difference was significant in colorectal cancer tissues, paracancerous tissues and distal cancer tissues. The positive rate of TLR-4 in cancer tissue, paracancerous tissue and distal cancer tissue was significantly lower than that in the control group (P < 0.05). Its expression level was gradually down-regulated. The positive rate of PGE-2 was 3.89% 3.19% and 1.01%, respectively. There was a significant difference among the three groups. The expression rate of P0.01U. TLR-4 in the PGE-2 positive group was 20 / 32, 62.50% and 50.00% respectively. In the negative group, the expression rate of PGE-2 in the TLR-4 positive group was 20 / 2483.330.The expression rate in the TLR-4 negative group was 12 / 16 / 75.00, and the positive rate of PGE-2 in the negative TLR-4 group was 20 / 2483.330.The expression rate of PGE-2 in the TLR-4 negative group was 12 / 16 / 75.00; In colorectal cancer, There were 20 cases of both positive expression and 4 cases of negative expression, the consistent rate of expression was 60.00. the analysis showed that the expression of both positive correlation, the difference was statistically significant (P 0.05). Conclusion the expression of TLR-4 in colorectal cancer tissues, paracancerous tissues and normal tissues of distal carcinoma decreased gradually. The farther away from the center of cancer tissue, the lower the positive area of TLR-4 expression was in colorectal cancer tissues. The level of expression in paracancerous tissues and normal tissues of distant cancer decreased gradually, and the farther away from the center of cancer tissue, The lower the positive area rate of TLR-4, the higher the expression of TLR-4 and PGE-2 in colorectal cancer tissues. TLR-4 may promote the malignant progression of colorectal cancer by up-regulating the expression of PGE-2.
