【摘要】：目的探討內(nèi)質(zhì)網(wǎng)應(yīng)激相關(guān)因子蛋白激酶R樣內(nèi)質(zhì)網(wǎng)調(diào)節(jié)激酶(PERK)和活化轉(zhuǎn)錄因子6(ATF6)在結(jié)直腸癌組織中的表達(dá)情況,分析PERK和ATF6在結(jié)腸癌發(fā)生發(fā)展中的作用。方法選擇手術(shù)切除結(jié)腸癌組織及距離病變組織5 cm以上正常組織,采用實(shí)時(shí)熒光定量PCR技術(shù)(RTPCR)檢測(cè)PERK和ATF6的mRNA表達(dá)情況,免疫組織化學(xué)法(IHC)、Western blot法檢測(cè)PERK和ATF6蛋白的表達(dá)情況,并結(jié)合臨床病理特征,分析其與結(jié)腸癌發(fā)生、發(fā)展之間的關(guān)系。結(jié)果腫瘤組織ATF6和PERK mRNA表達(dá)均較正常組織下調(diào)(P0.05)。IHC和Western blot結(jié)果顯示PERK和ATF6在結(jié)腸癌中的表達(dá)均顯著低于正常組織(P0.05)。PERK和ATF6主要定位于上皮細(xì)胞中。結(jié)論P(yáng)ERK和ATF6在結(jié)腸癌中的表達(dá)水平下降說明缺乏適當(dāng)?shù)膬?nèi)質(zhì)網(wǎng)應(yīng)激反應(yīng)可能在腫瘤的發(fā)生機(jī)制中起作用。
[Abstract]:Objective to investigate the expression of ER stress-related factor protein kinase R-like endoplasmic reticulum regulated kinase (ERK) and activated transcription factor 6 (ATF6) in colorectal carcinoma and to analyze the role of PERK and ATF6 in the carcinogenesis and development of colon cancer.Methods the tissues of colon cancer and normal tissues more than 5 cm away from the lesion were resected. The expression of PERK and ATF6 was detected by real-time fluorescence quantitative PCR and the expression of PERK and ATF6 protein was detected by immunohistochemistry.Combined with clinicopathological features, the relationship between colonic cancer and the occurrence and development of colon cancer was analyzed.Results the expression of ATF6 and PERK mRNA in tumor tissues was significantly lower than that in normal tissues. The results showed that the expression of PERK and ATF6 in colon cancer was significantly lower than that in normal tissues. P0.05, PERK and ATF6 were mainly localized in epithelial cells.Conclusion the decreased expression of PERK and ATF6 in colon cancer suggests that the lack of proper endoplasmic reticulum stress may play a role in the pathogenesis of cancer.
