【摘要】：目的:研究miR-126-5p在胃癌組織及胃癌旁組織的表達(dá),初步探討miR-126-5p與胃癌發(fā)生、發(fā)展之間的關(guān)系。方法:利用基因芯片技術(shù)檢測14例早癌患者的胃癌組織及其癌旁組織,進(jìn)行miRNAs的表達(dá)譜情況分析;收集42例胃癌組織及其癌旁組織標(biāo)本,利用實(shí)時熒光實(shí)時定量PCR(q RT-PCR)檢測miR-126-5p的表達(dá)情況,依據(jù)實(shí)驗(yàn)結(jié)果來初步探討在胃癌組織中miR-126-5p的表達(dá)情況與胃癌的產(chǎn)生、進(jìn)展及胃癌患者的臨床病理資料之間的相關(guān)性。結(jié)果:miR-126-5p在胃癌組織中的ΔCT值為5.440±1.650,在癌旁組織中ΔCT值為7.329±1.456,miR-126-5p在胃癌組織中表達(dá)上調(diào)的有34個,下調(diào)的有8個,miR-126-5p在胃癌組織中的表達(dá)是上調(diào)的,結(jié)果具有統(tǒng)計(jì)學(xué)意義。對癌組織及癌旁組織的ΔCT值做ROC曲線分析,得出ΔCT值ROC曲線下的面積是0.818,該指標(biāo)對胃癌診斷有一定的準(zhǔn)確性。所得到的最佳臨界值為7.125。當(dāng)ΔC T≤7.125時,其對胃癌診斷的靈敏性為88.10%。miR-126-5p相對表達(dá)量ΔCT與臨床病理資料分析,結(jié)果示P值大于0.05,miR-126-5p在胃癌組織中的表達(dá)與臨床病理資料無明顯相關(guān)性。結(jié)論:miR-126-5p在胃癌組織中是高表達(dá)的,對胃癌具有一定的診斷價(jià)值。
[Abstract]:Objective: to study the expression of miR-126-5p in gastric carcinoma and its adjacent tissues, and to explore the relationship between miR-126-5p and carcinogenesis and development of gastric cancer. The expression profile of miRNAs was analyzed and the expression of miR-126-5p was detected by real-time quantitative PCR(q RT-PCR in 42 cases of gastric carcinoma and its adjacent tissues. According to the results of the experiment, the expression of miR-126-5p and the production of gastric cancer were preliminarily studied. Results the 螖 CT value of 1: miR-126-5p was 5.440 鹵1.650 in gastric cancer tissue and 7.329 鹵1.456miR-126-5p in paracancerous tissue. Eight down-regulated expression of miR-126-5p was up-regulated in gastric cancer tissues, and the results were statistically significant. The 螖 CT values of cancer tissues and adjacent tissues were analyzed by ROC curve.
