食管鱗癌中差異表達的lncRNA的篩選及分析

【摘要】:目的:篩查及分析食管鱗癌中長鏈非編碼RNA(lncRNA)及mRNA的差異表達譜,以及這些差異lncRNAs的功能、互作分子。方法:通過對3對食管鱗癌/癌旁組織的基因芯片篩選及生物信息學(xué)分析,預(yù)測這些差異表達的lncRNAs潛在靶點、通路,及其與臨近編碼基因、轉(zhuǎn)錄因子的相關(guān)性。結(jié)果:按FC值≥2.0且P值≤0.05的標(biāo)準(zhǔn),共篩選得到癌及癌旁組織差異表達的lncRNA 182個,mRNA 108個,GO聚類和KEGG分析發(fā)現(xiàn)差異lncRNAs潛在靶標(biāo)基因與細(xì)胞周期調(diào)控、細(xì)胞分化、細(xì)胞外基質(zhì)等活性密切相關(guān),進一步生物分析得到lncRNA NONHSAT015779、NONHSAT08197、NONHSAT112918和NONHSAT115190等17個lncRNAs可能與食管鱗癌發(fā)生發(fā)展密切相關(guān)。結(jié)論:與癌旁組織相比,lncRNAs在食管鱗癌組織中顯示不同的表達模式,這些差異表達的lncRNAs可能在食管鱗癌發(fā)生和發(fā)展中發(fā)揮重要作用。
[Abstract]:Objective: to screen and analyze the differential expression profiles of long chain noncoding RNAs and mRNA in esophageal squamous cell carcinoma, and the function and interaction of these lncRNAs. Methods: three pairs of esophageal squamous cell carcinoma / paracancerous tissues were screened by microarray and analyzed by bioinformatics.
