Wnt5a在肥胖合并肝細(xì)胞癌中表達(dá)的初步研究

【摘要】:目的探討肥胖合并肝細(xì)胞癌中Wnt5a和β-連環(huán)蛋白(β-catenin)的表達(dá)情況。方法6周齡C57BL/6J小鼠分別給予普通飼料和高脂飼料喂養(yǎng)至14周齡(共8周),肥胖模型造模成功后采用尾靜脈注射法將Hepa1-6肝癌細(xì)胞株(對照組注射生理鹽水)分別接種于體質(zhì)量正常小鼠和肥胖小鼠,18周齡時(shí)處死小鼠,眼眶取血分析相關(guān)血清學(xué)指標(biāo),麻醉下取肝組織后HE染色觀察肝臟脂肪變性和肝細(xì)胞癌定植情況,免疫組化染色觀察Wnt5a和β-catenin的表達(dá)情況。結(jié)果 18周齡時(shí),高脂飼料喂養(yǎng)的肥胖小鼠肝臟細(xì)胞發(fā)生脂肪變性;接種肝癌細(xì)胞、體質(zhì)量正常和肥胖小鼠肝組織中均查見腫瘤細(xì)胞團(tuán),在接種肝癌細(xì)胞的肥胖小鼠中可見肝組織灶狀壞死。不同處理組小鼠血清總蛋白、天冬氨酸氨基轉(zhuǎn)移酶、谷丙轉(zhuǎn)氨酶、膽固醇和甲胎蛋白差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。免疫組化染色結(jié)果顯示,Wnt5a相對表達(dá)量為正常肝組織肥胖小鼠肝組織接種肝癌細(xì)胞后的體質(zhì)量正常小鼠肝組織接種肝癌細(xì)胞后的肥胖小鼠肝組織(P0.05);β-catenin的相對表達(dá)量為接種肝癌細(xì)胞后的肥胖小鼠肝組織肥胖小鼠肝組織和接種肝癌細(xì)胞后的體質(zhì)量正常小鼠肝組織正常肝組織(P0.05)。結(jié)論在肥胖小鼠中Wnt5a的表達(dá)下調(diào),β-catenin異常蓄積,可能與肝細(xì)胞癌的形成和進(jìn)展密切相關(guān)。
[Abstract]:Objective to investigate the expression of Wnt5a and 尾 -catenin in obese hepatocellular carcinoma (HCC). Methods C57BL / 6J mice aged 6 weeks were fed with normal diet and high fat diet to 14 weeks of age (8 weeks). Hepa1-6 hepatoma cells (control group) were inoculated with normal body weight mice and obese mice at the age of 18 weeks by tail vein injection, and the mice were killed at the age of 18 weeks. Serum samples were collected from the orbit, liver tissues were removed under anesthesia, liver steatosis and hepatocellular carcinoma colonization were observed by HE staining, and the expression of Wnt5a and 尾 -catenin were observed by immunohistochemical staining. Results at the age of 18 weeks, the expression of Wnt5a and 尾 -catenin were detected by HE staining. Fatty degeneration occurred in liver cells of obese mice fed with high fat diet and tumor cell mass was found in liver tissues of normal body mass and obese mice inoculated with hepatoma cells. Liver tissue necrosis was observed in obese mice inoculated with hepatoma cells. Serum total protein, aspartate aminotransferase, alanine aminotransferase were found in different treatment groups. The difference of cholesterol and alpha-fetoprotein was statistically significant (P 0.05). The results of immunohistochemical staining showed that the relative expression of Wnt5a was the same as that of normal liver tissue, obese mice, liver tissue inoculated with hepatoma cells, normal mice liver tissue inoculated with liver cancer fine. The relative expression of 尾 -catenin in the liver tissue of obese mice after inoculation with hepatoma cells and the normal liver tissue of mice inoculated with hepatoma cells was P0.050.Conclusion the relative expression of 尾 -catenin in the liver tissue of obese mice after inoculation of hepatoma cells and normal liver tissue of mice after inoculation of hepatoma cells is P0.05. The expression of Wnt5a was down-regulated and 尾 -catenin accumulated abnormally in mice. It may be closely related to the formation and progression of hepatocellular carcinoma.
