長(zhǎng)鏈非編碼RNA GAS5在子宮內(nèi)膜癌中的表達(dá)及對(duì)腫瘤侵襲的影響

【摘要】:目的探討長(zhǎng)鏈非編碼RNA GAS5在子宮內(nèi)膜癌組織中的表達(dá)及其對(duì)子宮內(nèi)膜癌HEC-1A細(xì)胞侵襲能力的影響。方法篩選2013年1月-2015年12月間于解放軍451醫(yī)院婦產(chǎn)科行手術(shù)切除的子宮內(nèi)膜癌患者的癌及癌旁組織50例,分析lncRNA-GAS5在組織中的表達(dá)水平及臨床意義;通過(guò)重組質(zhì)粒在人子宮內(nèi)膜癌HEC-1A細(xì)胞中過(guò)表達(dá)lncRNA-GAS5,采用Transwell侵襲小室模型檢測(cè)過(guò)表達(dá)lncRNA-GAS5后HEC-1A細(xì)胞侵襲能力變化,qRT-PCR檢測(cè)micro RNA-21的表達(dá)變化,qRT-PCR及Western blot檢測(cè)PTEN表達(dá)變化。結(jié)果 lncRNA-GAS5在子宮內(nèi)膜癌組織中表達(dá)水平低于對(duì)應(yīng)癌旁組織(t=4.013,P0.05),子宮內(nèi)膜癌組織中長(zhǎng)鏈非編碼核醣核酸生長(zhǎng)組織特異性轉(zhuǎn)錄體5(lncRNA-GAS5)低表達(dá)與腫瘤淋巴結(jié)轉(zhuǎn)移(t=2.389,P=0.017)及高FIGO分期(Ⅲ+Ⅳ期,t=2.131,P=0.021)差異有統(tǒng)計(jì)學(xué)意義;lncRNA-GAS5重組質(zhì)粒轉(zhuǎn)染HEC-1A細(xì)胞后可抑制細(xì)胞的侵襲能力(t=7.654,P=0.008);qRT-PCR檢測(cè)發(fā)現(xiàn)過(guò)表達(dá)lncRNA-GAS5后microRNA-21(miR-21)的表達(dá)水平升高(t=23.533,P0.05),qRT-PCR及Western blot結(jié)果證實(shí),過(guò)表達(dá)lncRNA-GAS5后可促進(jìn)PTEN的表達(dá)(P0.05)。結(jié)論 lncRNA-GAS5在子宮內(nèi)膜癌組織中表達(dá)降低,lncRNA-GAS5可能通過(guò)下調(diào)miR-21的表達(dá)進(jìn)而恢復(fù)PTEN的表達(dá)來(lái)抑制子宮內(nèi)膜癌轉(zhuǎn)移。
[Abstract]:Objective to investigate the expression of long chain noncoding RNA GAS5 in endometrial carcinoma and its effect on the invasiveness of endometrial carcinoma HEC-1A cells. Methods from January 2013 to December 2015, surgical excision of gynecology and obstetrics in 451 Hospital of PLA was performed. 50 cases of endometrial carcinoma and adjacent tissues, To analyze the expression level and clinical significance of lncRNA-GAS5 in tissues. The expression of micro RNA-21 in human endometrial carcinoma HEC-1A cells was detected by qRT-PCR and Western blot was used to detect the expression of PTEN by using Transwell invasive chamber model to detect the changes of invasion ability of HEC-1A cells after overexpression of lncRNA-GAS5. The expression level of lncRNA-GAS5 in endometrial carcinoma was lower than that in corresponding adjacent tissues (P 0.05). The low expression of long chain non-coding RNA specific transcripts (5lncRNA-GAS5) and lymph node metastasis in endometrial carcinoma tissues were lower than those in corresponding adjacent tissues (P 0.017) and high FIGO stage (P < 0.05). There was significant difference in the invasion ability of HEC-1A cells after transfection of recombinant plasmid LncRNA-GAS5. The expression level of microRNA-21mmiR-21 after lncRNA-GAS5 expression was detected by RT-PCR. The results of Western blot and microRNA-21mmiR-21 showed that the expression level of microRNA-21 mRNA was increased after transfection of GAS5 plasmid into HEC-1A cells. Overexpression of lncRNA-GAS5 can promote the expression of PTEN P0.05.Conclusion the decrease of lncRNA-GAS5 expression in endometrial carcinoma may inhibit the metastasis of endometrial carcinoma by down-regulating the expression of miR-21 and restoring the expression of PTEN.
