【摘要】：目的:探討miR-218調(diào)控SNX4蛋白對(duì)乳腺癌細(xì)胞增殖和侵襲能力的影響。方法:q PCR檢測(cè)乳腺癌組織和乳腺癌細(xì)胞株中miR-218的表達(dá)情況;分析miR-218的表達(dá)和乳腺癌臨床病理參數(shù)之間的關(guān)系;雙熒光素酶實(shí)驗(yàn)檢測(cè)miR-218和SNX4之間的關(guān)系;過(guò)表達(dá)miR-218后MTT實(shí)驗(yàn)和侵襲實(shí)驗(yàn)檢測(cè)乳腺癌細(xì)胞增殖和侵襲能力變化;過(guò)表達(dá)SNX4后MTT實(shí)驗(yàn)和侵襲實(shí)驗(yàn)檢測(cè)SNX4對(duì)乳腺癌細(xì)胞增殖和侵襲能力的恢復(fù)水平;裸鼠體外成瘤實(shí)驗(yàn)檢測(cè)miR-218對(duì)乳腺癌細(xì)胞株成瘤能力的影響。結(jié)果:miR-218在乳腺癌組織和MCF-7細(xì)胞株中表達(dá)水平較高,miR-218的表達(dá)與乳腺癌的病理分期相關(guān)以及淋巴結(jié)轉(zhuǎn)移情況有關(guān),SNX4可能是miR-218下游的作用靶點(diǎn);過(guò)表達(dá)miR-21可以抑制乳腺癌細(xì)胞的增殖侵襲能力,過(guò)表達(dá)SNX4后可以逆轉(zhuǎn)miR-218對(duì)乳腺癌細(xì)胞的抑制作用,過(guò)表達(dá)miR-218后可以抑制乳腺癌細(xì)胞株在裸鼠體內(nèi)的成瘤能力。結(jié)論:miR-218在乳腺癌中表達(dá)上調(diào),同時(shí)miR-218可以調(diào)控SNX4的表達(dá)而影響乳腺癌細(xì)胞增殖和侵襲能力。
[Abstract]:Objective: to investigate the effect of SNX4 protein regulated by miR-218 on the proliferation and invasion of breast cancer cells. Methods: the expression of miR-218 in breast cancer tissues and breast cancer cell lines was detected by PCR, and the relationship between the expression of miR-218 and the clinicopathological parameters of breast cancer was analyzed. The relationship between miR-218 and SNX4 was detected by double luciferase assay, and the changes of proliferation and invasion ability of breast cancer cells were detected by MTT assay and invasion assay after overexpression of miR-218. After overexpression of SNX4, MTT assay and invasion assay were used to detect the recovery of SNX4 to the proliferation and invasion of breast cancer cells. In vitro tumorigenesis assay in nude mice was used to detect the effect of miR-218 on the tumorigenic ability of breast cancer cell lines. Results the high expression of miR-218 in breast cancer tissues and MCF-7 cell lines was related to the pathological stage of breast cancer and lymph node metastasis. SNX4 may be the target of miR-218 downstream. Overexpression of miR-21 can inhibit the proliferation and invasion of breast cancer cells, and overexpression of SNX4 can reverse the inhibitory effect of miR-218 on breast cancer cells. Overexpression of miR-218 can inhibit the tumorigenic ability of breast cancer cell line in nude mice. Conclusion: miR-218 can up-regulate the expression of SNX4 in breast cancer, while miR-218 can regulate the expression of SNX4 and affect the proliferation and invasion of breast cancer cells.
