【摘要】：目的探討c-Jun基因啟動子多態(tài)性位點rs4646999與腸癌預后的相關性。方法采用Taq-Man探針法檢測436例腸癌患者rs4646999-673CT的基因型,通過Kaplan-Meier法和Cox回歸分析其生存曲線;采用免疫印跡分析多態(tài)性位點不同基因型的c-Jun蛋白表達水平。結果單因素分析表明攜帶TT基因型患者累積生存率顯著高于CT和CC基因型攜帶者,差異有統計學意義。多因素Cox回歸分析顯示分化程度、淋巴結轉移、遠處轉移、TNM分期和rs4646999位點基因型為預后因素,CT/CC基因型與腸癌較差的生存預后相關(P0.05)。蛋白表達分析發(fā)現CC基因型癌組織c-Jun蛋白表達升高,而TT基因型則相反。結論 c-Jun基因啟動子區(qū)域多態(tài)性位點rs4646999與腸癌生存預后相關,其作用機制可能是增強c-Jun蛋白表達導致腸癌發(fā)生發(fā)展。
[Abstract]:Objective to investigate the correlation between c-Jun gene promoter polymorphism rs4646999 and prognosis of colorectal cancer. Methods rs464699 was detected by Taq-Man probe method in 436 patients with colorectal cancer. The genotype of 9-673CT. The survival curve was analyzed by Kaplan-Meier method and Cox regression analysis. Results univariate analysis showed that the cumulative survival rate of patients with TT genotype was significantly higher than that of patients with CT and CC genotypes. Multivariate Cox regression analysis showed that the degree of differentiation, lymph node metastasis, distant metastasis, TNM staging and rs4646999 locus genotype were prognostic factors. CT/CC genotypes were associated with poor survival and prognosis of colorectal cancer. Protein expression analysis showed that c-Jun protein expression was increased in CC genotype carcinoma. Conclusion the polymorphism of c-Jun gene promoter region rs4646999 is associated with the survival and prognosis of colorectal cancer. Its mechanism may be to enhance the expression of c-Jun protein, leading to the carcinogenesis and development of colorectal cancer.
