非小細胞肺癌中人線粒體轉(zhuǎn)錄終止因子3蛋白的表達及臨床病理學(xué)意義

【摘要】:目的人線粒體轉(zhuǎn)錄終止因子3(h MTERF3)是線粒體基因表達及氧化磷酸化活性的負調(diào)控因子,其在臨床腫瘤樣本中的表達情況目前少見報道。文中旨在探討h MTERF3蛋白在非小細胞肺癌(NSCLC)組織中表達及其與臨床病理因素之間的關(guān)系,揭示h MTERF3的表達在NSCLC中的臨床病理學(xué)意義。方法實驗材料為大理市第一人民醫(yī)院病理科2008年至2010年存檔蠟塊,包括65例NSCLC組織和32例癌旁正常肺組織。采用免疫組織化學(xué)SP法檢測NSCLC組織和癌旁正常肺組織中h MTERF3的表達情況,采用Kaplan-Meier法計算生存率,Cox比例風(fēng)險模型分析影響NSCLC患者預(yù)后的危險因素。結(jié)果 65例NSCLC組織中,h MTERF3陽性表達31例(47.69%)。h MTERF3高表達的患者術(shù)后生存時間顯著低于h MTERF3低表達的患者[(30.09±3.35)月vs(57.61±7.16)月,P0.05]。h MTERF3陽性表達[HR=3.302,95%CI:1.598~6.905]和淋巴結(jié)轉(zhuǎn)移[HR=4.052,95%CI:1.212~12.398]是影響NSCLC患者預(yù)后的危險因素。結(jié)論 h MTERF3蛋白在NSCLC中的表達水平顯著增高,并且h MTERF3陽性表達和淋巴結(jié)轉(zhuǎn)移患者生存率會降低。提示h MTERF3可成為肺癌細胞轉(zhuǎn)移的標(biāo)志物,用于預(yù)測NSCLC的預(yù)后。
[Abstract]:Objective Human mitochondrial transcriptional termination factor (3hMTERF3) is a negative regulator of mitochondrial gene expression and oxidative phosphorylation activity. The expression of h MTERF3 protein was rarely reported in clinical tumor samples. The purpose of this paper is to investigate the expression of h MTERF3 protein in non small cell lung cancer (NSCLC). Expression in tissues and its relationship with clinicopathological factors. To reveal the clinicopathological significance of h MTERF3 expression in NSCLC. Methods the experimental materials were paraffin pieces from 2008 to 2010 in Department of Pathology of the first people's Hospital of Dali. The expression of h MTERF3 was detected by immunohistochemical SP method in 65 cases of NSCLC and 32 cases of adjacent normal lung tissues. The survival rate was calculated by Kaplan-Meier method and Cox proportional risk model was used to analyze the risk factors affecting the prognosis of NSCLC patients. Results in 65 cases of NSCLC tissues. H MTERF3 positive expression in 31 patients with high expression of h MTERF3 was significantly lower than that in patients with low expression of h MTERF3. [Vs(57.61 鹵7.16 months (30.09 鹵3.35) months (P0.05). H MTERF3 positive expression. [HRN 3.302 / 95 CI: 1.598 / 6.905] and lymph node metastasis. [HR=4.052. 95 CI: 1.212 ~ 12.398] is a risk factor for the prognosis of patients with NSCLC. Conclusion the expression of h MTERF3 protein in NSCLC is significantly increased. The positive expression of h MTERF3 and survival rate of patients with lymph node metastasis decreased, suggesting that h MTERF3 may be a marker of lung cancer cell metastasis and can be used to predict the prognosis of NSCLC.
