DLL4和CD105在卵巢癌中的表達(dá)及臨床意義

【摘要】:目的:通過檢測(cè)卵巢癌及正常卵巢組織中Notch-Delta樣配體(DLL4)和CD105表達(dá),同時(shí)測(cè)定這兩組患者血清中CD105的濃度,分析Notch-Delta樣配體(DLL4)和CD105在卵巢癌中的意義、它們之間的聯(lián)系及與卵巢癌患者的臨床病理參數(shù)的關(guān)系。方法:選取來自臨沂市中心醫(yī)院婦科2015年11月-2017年2月期間,自術(shù)中切除的65例卵巢癌標(biāo)本,同時(shí)間段手術(shù)切除的18例正常卵巢組織。所有標(biāo)本需符合以下標(biāo)準(zhǔn):1、所有患者無其它惡性腫瘤病史;2、患者病歷資料完整,且附有2名病理科醫(yī)師診斷的病理報(bào)告單;3、術(shù)前未進(jìn)行放化療、激素治療等;應(yīng)用免疫組化,懫用S-P法,在卵巢癌及正常卵巢組織中分別測(cè)定DLL4的表達(dá)及CD105標(biāo)記的微血管密度(CD105-MVD)。同時(shí)應(yīng)用酶聯(lián)免疫吸附法(ELISA)檢測(cè)上述所選患者的血清CD105濃度。結(jié)果:1、DLL4在卵巢癌中表達(dá)陽(yáng)性,大部分定位于細(xì)胞核,呈棕黃色細(xì)顆粒,而在正常卵巢組織中基本無表達(dá)或低表達(dá),DLL4的陽(yáng)性表達(dá)率在卵巢癌患者中為78.5%,在正常卵巢組織中的陽(yáng)性表達(dá)率為11.1%。2、卵巢癌中,DLL4的表達(dá)與患者的FIGO分期、卵巢癌的分化程度、淋巴結(jié)有無轉(zhuǎn)移有相關(guān)(P0.01),與卵巢癌的組織學(xué)類型、患者年齡無相關(guān)(P0.05)。3、CD105在卵巢癌中的表達(dá)為陽(yáng)性,主要定位于細(xì)胞漿、細(xì)胞膜,在正常卵巢組織中基本無表達(dá)或低表達(dá)。CD105-MVD在卵巢癌患者為22.58±7.34、在正常卵巢中是0.87±0.19。4、卵巢癌組織CD105與卵巢癌的分化程度、有無淋巴結(jié)轉(zhuǎn)移、FIGO分期有相關(guān)性(P0.05),和卵巢癌組織學(xué)類型、患者年齡無相關(guān)性(P0.05)。5、卵巢癌血清中CD105濃度(136.8642±26.6727)明顯高于正;颊哐逯械臐舛(26.6727±1.4111)。6、血清中CD105與卵巢癌的分化程度、有無淋巴結(jié)轉(zhuǎn)移、FIGO分期有相關(guān)性(P0.05),和卵巢癌組織學(xué)類型、患者年齡無相關(guān)性(P0.05)。7、卵巢癌組織中DLL4和CD105表達(dá)的關(guān)呈正相關(guān)(r=0.818,P0.001)。8、卵巢癌組織中CD105-MVD與血清CD105呈正相關(guān)(r=0.692,P0.001)。9、卵巢癌組織中DLL4與血清CD105的濃度呈正相關(guān)(r=0.690,P0.001)。結(jié)論:DLL4、CD105在腫瘤新生血管中起重要作用。隨著卵巢癌的臨床進(jìn)展,組織中的DLL4、CD105及血清CD105都呈現(xiàn)不斷增高的趨勢(shì),且呈正相關(guān),提示在卵巢癌中,DLL4可使血管密度增加,從而參與卵巢癌的發(fā)生、發(fā)展。DLL4、CD105可作為判斷卵巢癌侵襲、轉(zhuǎn)移的重要指標(biāo),并為抗血管治療提供依據(jù)。
[Abstract]:Objective: to detect the expression of Notch-Delta like ligand DLL4 (DLL4) and CD105 in ovarian cancer and normal ovarian tissues, and to determine the serum CD105 concentration in these two groups at the same time. To analyze the significance of Notch-Delta like ligand DLL4) and CD105 in ovarian cancer. The relationship between them and the clinicopathological parameters of patients with ovarian cancer. Methods: from November 2015 to February 2017, the gynecology department of Linyi central hospital was selected. 65 cases of ovarian cancer were resected during operation and 18 cases of normal ovarian tissue were resected at the same time. All specimens should meet the following standard: 1. All patients had no history of malignant tumor. (2) the patient's medical records were complete, and two pathologists' pathological reports were attached. 3, no preoperative chemotherapy, chemotherapy, hormone therapy, and so on; Immunohistochemistry and S-P method were used. The expression of DLL4 and the microvessel density of CD105-MVD labeled by CD105 were measured in ovarian cancer and normal ovarian tissues, respectively. The serum CD105 concentration of the above selected patients was measured. Results: 1. The expression of DLL4 was positive in ovarian cancer, most of which were located in the nucleus and showed brown and fine granules, but there was no or low expression in normal ovarian tissues. The positive expression rate of DLL4 was 78.5 in ovarian cancer and 11.1. 2 in normal ovarian tissue. The expression of DLL4 in ovarian cancer and the FIGO stage of ovarian cancer. The degree of differentiation of ovarian cancer, lymph node metastasis has a correlation with P0.01, and the histological type of ovarian cancer, the patient age is not related to P0.05. 3. The expression of CD105 was positive in ovarian cancer, mainly located in cytoplasm and cell membrane. The expression of CD105-MVD was 22.58 鹵7.34 in ovarian cancer and 0.87 鹵0.19.4 in normal ovary. There was a correlation between CD105 and differentiation degree of ovarian cancer, FIGO stage with or without lymph node metastasis, and histological type of ovarian cancer. There was no correlation between age and age (P 0.05). The serum CD105 concentration of ovarian cancer was 136.8642 鹵26.6727, which was significantly higher than that of normal patients (26.6727 鹵1.4111). Serum CD105 was correlated with the differentiation degree of ovarian cancer, FIGO stage with or without lymph node metastasis, and histological type of ovarian cancer. The expression of DLL4 and CD105 in ovarian cancer tissues was positively correlated with the expression of P0. 818 and P0. 001. 8 in patients with ovarian cancer, there was no correlation between age and age (P0. 05. 7, P 0. 05. 7). There was a positive correlation between CD105-MVD and serum CD105 in ovarian cancer tissues. There was a positive correlation between the concentration of DLL4 and serum CD105 in ovarian cancer tissues. CD105 plays an important role in tumor neovascularization. With the clinical progress of ovarian cancer, both DLL4CD105 and serum CD105 have a positive correlation with each other. It is suggested that DLL4 can increase the density of blood vessels and participate in the development of ovarian cancer. DLL4CD105 can be used as an important index to judge the invasion and metastasis of ovarian cancer. And to provide the basis for anti-vascular therapy.
