【摘要】:目的探究穿膜肽修飾的納米顆粒介導(dǎo)的光動(dòng)力學(xué)療法(PDT)對(duì)胰腺癌SW1990細(xì)胞的殺傷作用。方法將胰腺癌SW1990細(xì)胞分成4組,不光照不含光敏劑的細(xì)胞作為空白對(duì)照組,與細(xì)胞共孵育的游離Ce6組,NP/Ce6組及TAT-NP/Ce6組。首先分別制備出納米顆粒TAT-NP/Ce6和NP/Ce6。利用流式細(xì)胞儀(FACS)檢測(cè)各組被SW1990細(xì)胞攝取的情況;熒光顯微鏡觀察各組光照下在細(xì)胞內(nèi)產(chǎn)生活性氧的情況;最后用MTT或活死染色法檢測(cè)光照下各組對(duì)細(xì)胞的殺傷效果。結(jié)果 FACS結(jié)果顯示各組分別被細(xì)胞攝取4 h后,TAT-NP/Ce6組胞內(nèi)熒光顯著高于NP/Ce6組(P0.001),同時(shí)NP/Ce6組明顯強(qiáng)于free Ce6組(P0.001)。熒光顯微鏡顯示光照下納米顆粒TAT-NP/Ce6組在細(xì)胞內(nèi)產(chǎn)生活性氧明顯強(qiáng)于其他各組(P0.001)。MTT法顯示在每一Ce6濃度下光照時(shí),TAT-NP/Ce6組對(duì)細(xì)胞的殺傷效果明顯強(qiáng)于NP/Ce6(P0.01),Ce6濃度為0.5μg/ml時(shí),活死細(xì)胞染色法同樣證明了對(duì)細(xì)胞的這種殺傷效果。結(jié)論TAT肽修飾的納米顆粒能有效增加胰腺癌細(xì)胞對(duì)其攝取,其介導(dǎo)的PDT殺傷胰腺癌細(xì)胞效果明顯增強(qiáng)。
[Abstract]:Objective to investigate the killing effect of transmembrane peptide modified nanoparticles (PDT) on pancreatic cancer SW1990 cells. Methods Pancreatic carcinoma SW1990 cells were divided into 4 groups. The cells without photoperiod and without Guang Min were used as the blank control group and the free Ce6 group co-incubated with the cells. In NP/Ce6 group and TAT-NP/Ce6 group, the nanoparticles TAT-NP/Ce6 and NPP / Ce6 were prepared, respectively. Flow cytometry (FCM) was used. The uptake of SW1990 cells in each group was detected. Fluorescence microscope was used to observe the production of reactive oxygen species (Ros) in the cells under light. Finally, MTT or live death staining was used to detect the killing effect of each group under light. Results the FACS results showed that the cells in each group were ingested for 4 hours. The intracellular fluorescence in TAT-NP/Ce6 group was significantly higher than that in NP/Ce6 group (P 0.001). At the same time, NP/Ce6 group was significantly stronger than free Ce6 group (P0.001). Fluorescence microscope showed that the reactive oxygen species (Ros) production in the TAT-NP/Ce6 group was significantly stronger than that in the other groups (P0.001). MTT method was used to show the light intensity at each Ce6 concentration. The killing effect of TAT-NP/Ce6 group was significantly better than that of NP- / Ce6P0.01Ce _ 6 concentration of 0.5 渭 g / ml. TAT peptide modified nanoparticles can effectively increase the uptake of pancreatic cancer cells. The effect of PDT mediated killing pancreatic cancer cells was significantly enhanced.
