電針干預(yù)骨癌痛大鼠的實(shí)驗(yàn)研究

【摘要】:目的:通過電針刺激乳腺癌骨轉(zhuǎn)移大鼠,觀察電針對骨癌痛的影響,討論其可行性,并探討電針治療骨癌痛的作用機(jī)理,以指導(dǎo)臨床運(yùn)用 方法:1、將24只體重180-220g的SD雌性大鼠,隨機(jī)分為三組:空白組、假手術(shù)組、模型組。將Walker256大鼠乳腺癌細(xì)胞經(jīng)腹水傳代后(1×105cells)接種至SD大鼠左側(cè)后肢脛骨骨髓腔內(nèi)制備模型。與空白組、假手術(shù)組比較判斷造模成敗。2、將40只體重180-220g的SD雌性大鼠接種Walker256大鼠乳腺癌細(xì)胞造模后隨機(jī)分為5組:空白組、模型組、電針組、藥物組、針?biāo)幗M,每組8只,分別干預(yù)后,觀察各組大鼠行為學(xué)、形態(tài)學(xué)及利用免疫組化法和Western-blot法觀察抗酒石酸酸性磷酸酶(Tartrate resistant acid phosphatase,TRAP)、膠質(zhì)纖維酸性蛋白(Glial Fibrillary Acidic Protein,GFAP)的表達(dá)。 結(jié)果: ].大鼠接種Walker256乳腺癌細(xì)胞后,X線攝片可見術(shù)側(cè)脛骨在接種后8天、14天和21天分別出現(xiàn)不同程度的骨質(zhì)損傷和破壞雙側(cè)后肢熱痛過敏,鼠尾出現(xiàn)機(jī)械性觸痛過敏;術(shù)側(cè)脛骨HE染色在接種后21天可見大量的腫瘤細(xì)胞; 2.大鼠接種后21天脛骨重酒石酸酸性磷酸酶(TRAP)染色表達(dá)上調(diào),同時激活脊髓膠質(zhì)細(xì)胞的標(biāo)志物GFAP; 3.應(yīng)用電針、唑來膦酸及電針+唑來膦酸治療均能夠使上調(diào)的TRAP表達(dá)下降,同時抑制被激活的GFAP; 4.電針+唑來膦酸對骨癌痛的治療作用明顯優(yōu)于單純電針或唑來膦酸的作用。 結(jié)論: 1.利用Walker256乳腺癌細(xì)胞能夠成功制出大鼠乳腺癌骨轉(zhuǎn)移模型 2.應(yīng)用電針、藥物及針?biāo)幗Y(jié)合均可緩解大鼠骨癌痛,且針?biāo)幗Y(jié)合療效明顯優(yōu)于單純電針或藥物治療; 3.電針對癌癥骨轉(zhuǎn)移疼痛具有治療作用,而且電針結(jié)合藥物能夠提高鎮(zhèn)痛效果。
[Abstract]:Objective: to observe the effect of electroacupuncture on bone cancer pain, discuss its feasibility and explore the mechanism of electroacupuncture in treating bone cancer pain in rats with bone metastases of breast cancer. Methods Twenty four female SD rats weighing 180-220 g were randomly divided into three groups: blank group, sham operation group and model group. Walker256 rat breast cancer cells were inoculated into the left hindlimb tibial medullary cavity of SD rats after ascites passage and 1 脳 105 cells were inoculated into the tibial medullary cavity of the left hind limb to make the model. 40 SD female rats weighing 180-220 g were randomly divided into 5 groups: blank group, model group, electroacupuncture group, drug group, acupuncture group, and acupuncture group. After intervention, the behavior, morphology and the expression of tartrate resistant acid phosphatase, glial fibrillary acidic protein (Glial Fibrillary Acidic protein Western-blot) were observed by immunohistochemical and Western-blot methods. Results: ]. After Walker256 breast cancer cells were inoculated, the tibia of the operative side appeared different degrees of bone injury and hyperthermia on the 14th and 21st days after inoculation, and the mechanical tenderness was found in the tail of the rat. A large number of tumor cells were observed 21 days after inoculation with HE staining in the operative tibia. 2. The expression of tibial heavy tartrate acid phosphatase (TRAP) was up-regulated 21 days after inoculation, and the marker of glial cells in spinal cord was activated. 3. Electroacupuncture, zoledronic acid and electroacupuncture zoledronic acid could decrease the up-regulated expression of TRAP and inhibit the activated GFAPs. 4. The therapeutic effect of electroacupuncture zoledronic acid on bone cancer pain is better than that of electroacupuncture alone or zoledronic acid alone. Conclusion: 1. Bone Metastasis Model of Breast Cancer in Rats can be successfully established by using Walker256 Breast Cancer cells 2. The combination of electroacupuncture, medicine and acupuncture can relieve the pain of bone cancer in rats, and the curative effect of acupuncture combined with medicine is better than that of simple electroacupuncture or drug therapy. 3. Electroacupuncture can treat cancer bone metastases pain, and electroacupuncture combined with drugs can improve analgesic effect.
