【摘要】：探討熊去氧膽酸(UDCA)和"一貫煎"在原發(fā)性膽汁性肝硬化小鼠中的療效。對(duì)polyI∶C注射的C57BL/6雌性小鼠分別給予UDCA、"一貫煎"和兩種藥物聯(lián)合治療,分析不同藥物治療組血清ALP、-γGT的變化,實(shí)時(shí)熒光定量RT-PCR方法檢測(cè)脾臟單個(gè)核細(xì)胞BCMA mRNA的表達(dá),并分析肝門(mén)管區(qū)單個(gè)核細(xì)胞浸潤(rùn)率。結(jié)果表明,PBC模型鼠肝門(mén)管區(qū)單個(gè)核細(xì)胞浸潤(rùn)率、脾臟單個(gè)核細(xì)胞BCMA mRNA的表達(dá)較正常小鼠明顯增加(P0.05);不同治療組較模型對(duì)照組ALP、-γGT顯著下降(P0.05),"一貫煎"或聯(lián)合治療還能減少肝門(mén)管區(qū)單個(gè)核細(xì)胞浸潤(rùn)率、下調(diào)脾臟單個(gè)核細(xì)胞BCMAmRNA和ALP。"一貫煎"或聯(lián)合熊去氧膽酸治療PBC小鼠療效更好,為以后中西醫(yī)聯(lián)合治療PBC提供了理論依據(jù)和技術(shù)支持。
[Abstract]:To investigate the efficacy of ursodeoxycholic acid (UDCA) and "Yijian" in primary biliary cirrhosis mice. The female C57BL/6 mice injected with polyI:C were treated with UDCA, "Yijian" and two drugs, respectively. The changes of serum ALP- 緯 GT in different drug treatment groups were analyzed, and the expression of BCMA mRNA in spleen mononuclear cells was detected by real-time fluorescence quantitative RT-PCR. The infiltration rate of mononuclear cells in hepatic hilar area was analyzed. The results showed that the infiltration rate of mononuclear cells in hepatic hilar area of PBC model rats, The expression of BCMA mRNA in splenic mononuclear cells was significantly higher than that in normal mice, and the expression of BCMA mRNA in spleen mononuclear cells was significantly decreased in different treatment groups than that in the model control group. "Yijianjian" or combined treatment could also reduce the infiltration rate of mononuclear cells in the hilar area of the liver and down-regulate the BCMAmRNA and ALP of splenic mononuclear cells. " Yijianjian "or ursodeoxycholic acid combined with ursodeoxycholic acid is more effective in treating PBC mice, which provides theoretical basis and technical support for the later treatment of PBC by combination of traditional Chinese and western medicine."
