黃芪多糖降低A549/DDP肺腺癌細(xì)胞順鉑耐藥機(jī)制研究

目的:研究黃芪多糖對(duì)A549/DDP肺腺癌細(xì)胞順鉑耐藥的影響及可能機(jī)制。方法:將A549/DDP順鉑耐藥肺腺癌細(xì)胞分為CON組(未行黃芪多糖及順鉑處理)、APS組(僅用黃芪多糖處理)、DDP組(僅用順鉑處理)及A+D組(黃芪多糖聯(lián)合順鉑處理),比較四組細(xì)胞增殖、細(xì)胞周期及凋亡率和耐藥基因表達(dá)的差異。結(jié)果:A+D組細(xì)胞d1-d7吸光度、S期、G2/M期和PI均顯著低于CON組、APS組和DDP組細(xì)胞(均P<0.05),G0/G1期和凋亡率均顯著高于CON組、APS組和DDP組細(xì)胞(均P<0.05);APS組和A+D組細(xì)胞MDR1和MRP基因mRNA表達(dá)無明顯差異(均P>0.05),且均顯著低于CON組和DDP組(均P<0.05)。結(jié)論:黃芪多糖可顯著改善A549/DDP肺腺癌細(xì)胞對(duì)順鉑耐藥性,可能與黃芪多糖顯著降低MDR1和MRP有關(guān)。
