蒙古蟲實(shí)和垂盆草的化學(xué)成分及其抗炎作用研究

本論文對(duì)沙生植物蒙古蟲實(shí)(Corispermum mongolicum Iljin)和傳統(tǒng)藥用植物垂盆草(Sedum sarmentosum Bunge)的化學(xué)成分及其抗炎活性進(jìn)行了深入研究。蒙古蟲實(shí)是一種典型的沙生灌木,是藜科蟲實(shí)屬的一年生草本植物。蒙古蟲實(shí)廣泛分布于中國(guó)沙漠地區(qū),主要用作防風(fēng)固林,并具有較高的飼用價(jià)值。然而,該植物的提取物及單體化合物的藥理活性未見報(bào)道。為了明確其活性成分,本研究利用各種柱層析的方法,對(duì)蒙古蟲實(shí)乙醇提取物的石油醚萃取部位、乙酸乙酯萃取部位及正丁醇萃取部位分別進(jìn)行了分離及純化,共得到38個(gè)化合物,其中包 括 1 個(gè) 新 的 木 脂 素 類 化 合 物:3f→3a:6f→2a-[(3R,4R)-4',6-dihydroxy-3',5,7-trimethoxy-4,10-cycloligna-1-en-2a,3-dicarbonyl]-6g-(p-courmaroyl)sucrose(1),2 個(gè)新的降碳倍半萜類化合物:(-)-(3R)-hydroxysulfonyloxy-β-ionone(13),(3S,6S,9R)-3,9,13-trihydroxymegastigman-4-ene-3-O-β-D-glucopyranoside(14),其余 35個(gè)已知化合 物 分 別 為:trigonotin C(2),acanthoside B(3),(-)-pinoresinol-4-O-p-D-glucopyranoside(4),methyl rosmarinate(5),(R)-(+)-oresbiusin B(6),lariciresinol-4'-O-β-D-glucopyranoside(7),lariciresinol-4-O-β-D-glucopyranoside(8),isolariciresinol-4-O-β-D-glucopyranoside(9),(7S,8R)-dehydrodiconifervl alcohol-4-O-β-D-glucopyranoside(10)和sargentodoside D(12)的混合 物,(7S,8R)-dehydrodiconiferyl alcohol-9'-O-β-D-glucopyranoside(11),byzantionoside B(15),icariside B2(16),3α-hydroxysulfonyloxy-5α,6αα-epoxy-7-megastigmen-9-one(17),(-)-(R)-dehydrovomifoliol(18),6'-O-palmitoylsitosterol-3-O-β-D-glucoside(19),daucosterol(20),β-sitosterol(21),blazein(22),(3β,4α)-3-hydroxy-24-methylene-9,19-cyclolanostan-28-oic acid(23),stigmasta-3,6-dione(24),stigmasta-4-ene-3,6-dione(25),stigmasta-4,22-diene-3,6-dione(26),astragalin(27),hirsutrin(28),indole-3-carboxaldehyde(29),(+)-(S)-oresbiusin A(30),benzyl-O-β-D-glucopyranoside(31),2-phenylethyl-O-β-D-glucopyranoside(32),(Z)-3-hexenyl β-D-glucopyranoside(33),α-amyrin(34),cassipourol(35),1-O-β-D-glucopyranosyl-(2S,3S,4R,8E/Z)-2-[(2'R)-2'-hydroxypalmitoylamino]-8-octa decene-l,3.4-triol(36),soya-cercbrosidel(37),panaxcerol C(38)。其中,化合物 2、4-12、15、18-19、22-26、29-30、32-35和37是首次從藜科中分離得到。垂盆草為景天科景天屬多年生草本植物,廣泛分布在我國(guó)山區(qū),比如長(zhǎng)白山地區(qū)。其顆粒劑在臨床用于治療肝炎已有近40年的歷史。研究表明垂盆草具有提高免疫力、保護(hù)肝臟、抗癌、抗菌、抗炎等特性。國(guó)內(nèi)外文獻(xiàn)報(bào)道垂盆草提取物具有抗炎活性,但從該植物中提取的單體化合物的抗炎活性研究尚未見報(bào)道。為了進(jìn)一步研究垂盆草的抗炎活性物質(zhì),本研究利用各種色譜分離技術(shù),對(duì)垂盆草的乙酸乙酯萃取部位及正丁醇萃取部位分別進(jìn)行了系統(tǒng)分離和純化,并通過(guò)波譜學(xué)分析方法共鑒定出33個(gè)化合物(39-71),其中包括1個(gè)新的黃酮類化合物limocitrin-3-O-α-L-arabinopyranoside(39),其余 32 個(gè)已知化合物分別是limocitrin-3-O-β-D-glucopyranoside(40),myrsinionoside A(41),sarmentol A(42),(3S,5R,6S,9R)-megastigmane-3,9-diol(43),sarmentol C(44),neosarmentol Ⅲ(45),vomifoliol(46),sarmentol D(47),(+)-(S)-dehydrovomifoliol(48),actinidioionoside(49),(3S,5R,6R,7E,9S)-3,5,6,9-tetrahydroxymegastigman-7-ene(50),sedumoside Ⅰ(51),myrsinionoside D(52),sedumosides E2(53),sedumosides E1(54),sedumosides A2(55)和 sedumosides A3(56)的混合物,(+)-pinoresinol(57),(+)-pinoresinol di-O-β-D-glucopyranoside(58),(+)-epipinoresinol(59),(+)-isolariciresinol(60),burselignan(61),α-amyrone(62),δ-amyrin(63),3-epi-δ-amyrin(64),(3β,4α,5α)-4,14-dimethyl-cholest-9(11)-en-3-ol(65),20-hydroxyecdysone(66),stigmast-5-ene-7α-methoxy-3β-ol(67),methyl-p-hydroxy cinnamate(68),homovanillyl alcohol(69),2-methoxy-4-(2-propenyl)phenyl β-D-glucopyranoside(70),octa-1-en-3-yl α-L-arabinopyranosyl(1→6)-β-D-glucopyranoside(71);衔46、48-50、58-59、61、65、67-68是首次從景天科中分離得到。本實(shí)驗(yàn)進(jìn)一步測(cè)定了單體化合物(1-71)的抗炎作用,采用MTT法檢測(cè)細(xì)胞存活率、Griess和ELISA方法檢測(cè)化合物對(duì)LPS誘導(dǎo)的RAW 264.7細(xì)胞上清液中炎癥介質(zhì)NO,TNF-α和IL-6的釋放的影響。結(jié)果顯示,化合物5,6,14,27,28 和 41-50 在濃度范圍為 10,30,100 μM 時(shí),對(duì) NO,TNF-α 和 IL-6 均具有明顯的抑制作用,其半數(shù)抑制濃度為34.37-96.21 μM。
