模擬失重降低大鼠心肌細(xì)胞對(duì)異丙腎上腺素的反應(yīng)性

【摘要】:本文旨在研究模擬失重對(duì)大鼠單個(gè)心肌細(xì)胞無(wú)負(fù)荷收縮功能的影響以及對(duì)異丙腎上腺素(isoproterenol,ISO)反應(yīng)性的變化。采用大鼠尾部懸吊法在地面模擬失重狀態(tài),4周后以膠原酶Ⅰ消化分離心肌細(xì)胞,分別對(duì)左、右兩心室心肌細(xì)胞進(jìn)行收縮功能測(cè)量。結(jié)果顯示,懸吊4周大鼠(懸吊組)左、右心室心肌細(xì)胞的長(zhǎng)度和寬度與正常大鼠(對(duì)照組)相比均無(wú)顯著差異。隨刺激頻率增加,對(duì)照組與懸吊組大鼠心肌細(xì)胞縮短幅值均逐步增加。在1.0、2.0與4.0 Hz刺激下,對(duì)照組大鼠左心室心肌細(xì)胞縮短幅值分別為(8.50±1.26)%、(9.00±1.38)%與(9.23±1.83)%,右心室心肌細(xì)胞縮短幅值分別為(9.80±2.48)%、(10.03±2.48)%與(10.28±2.27)%:與對(duì)照組大鼠相比,在1.0與2.0 Hz刺激下,懸吊組大鼠左心室心肌細(xì)胞無(wú)負(fù)荷縮短幅值分別降低12.2%、10.9%(P0.05),右心室則分別降低16.5%、16.3%(P0.05);但是在4.0 Hz刺激下卻無(wú)顯著性改變。與同一頻率刺激下的對(duì)照組大鼠相比,懸吊組大鼠左、右心室心肌細(xì)胞達(dá)到縮短峰值的時(shí)程(time to peak shortening,TPS)明顯縮短(P0.05);而從縮短峰值至75%舒張的時(shí)程(TR_(75))則明顯延長(zhǎng)(P0.05)。在各刺激頻率下,懸吊組大鼠左、右心室心肌細(xì)胞縮短(+dL/dt_(max))與舒張(-dL/dt_(max))速度均未發(fā)生明顯改變。用1、5、10 nmol/L ISO灌流達(dá)穩(wěn)態(tài)水平后,對(duì)照組大鼠心肌細(xì)胞縮短幅值分別增加了(10.63±0.83)%、(35.06±5.22)%和(71.64±6.83)%;而懸吊組大鼠心肌細(xì)胞縮短幅值僅增加(5.75±0.76)%、(23.97±4.50)%和(26.38±8.13)%,均有顯著性差異(P0.05,P0.01)。用10、50、100 nmol/L forskolin灌流達(dá)穩(wěn)定水平后,對(duì)照組大鼠心肌細(xì)胞縮短幅值分別增加了(3.04±0.27)%、(9.81±2.66)%、(20.20±3.47)%;而懸吊組大鼠心肌細(xì)胞縮短幅值僅增加了(1.42±0.53)%、(3.83±1.71)%、(5.49±4.08)%,均有顯著性差異(P0.05)。以上結(jié)果表明,模擬失重4周降低大鼠心肌細(xì)胞無(wú)負(fù)荷縮短幅值以及對(duì)ISO的反應(yīng)性。
