不同強(qiáng)度推拉動(dòng)作對(duì)大鼠海馬神經(jīng)元bFGF表達(dá)的影響

【摘要】:目的 :采用動(dòng)物離心機(jī)建立模擬推拉動(dòng)作的動(dòng)物模型 ,探討“推拉效應(yīng)”對(duì)大鼠海馬神經(jīng)元bFGF表達(dá)的影響 .方法 :健康雄性Wistar大鼠 4 0只 ,隨機(jī)分為 1 0組 (對(duì)照組 ,三種強(qiáng)度暴露后的 30min,6h ,2 4h組 ) ,每組 4只 ,灌注固定 ,取大腦半球 ,切片后應(yīng)用免疫組織化學(xué)染色及圖像分析技術(shù)對(duì)大鼠海馬各區(qū)神經(jīng)元堿性成纖維細(xì)胞生長(zhǎng)因子 (bFGF)表達(dá)進(jìn)行檢測(cè) .結(jié)果 :對(duì)照組和± 2Gz暴露后的 30min組bFGF呈弱陽(yáng)性表達(dá) ;而± 6Gz和± 8Gz暴露后 30min的實(shí)驗(yàn)組可見(jiàn)大鼠海馬神經(jīng)元細(xì)胞bFGF表達(dá)開(kāi)始增加 ,6h實(shí)驗(yàn)組表達(dá)明顯增強(qiáng) ,2 4h后表達(dá)下降但仍較正常對(duì)照組高 .與對(duì)照組比較 ,差異有顯著性 (P 0 .0 5 ) .結(jié)論 :中、高強(qiáng)度的推拉動(dòng)作可引起大鼠海馬神經(jīng)元bFGF表達(dá)增加
[Abstract]:Aim: to establish an animal model with animal centrifuge to simulate push-pull action and to investigate the effect of "push-pull effect" on the expression of bFGF in hippocampal neurons of rats. Methods: forty healthy male Wistar rats were randomly divided into 10 groups (control group, 30 min after exposure to three intensities for 6 h and 24 h), each group (4 rats) were infused and fixed, and the hemispheres were taken. The expression of basic fibroblast growth factor (bFGF) in hippocampal neurons of rats was detected by immunohistochemical staining and image analysis. Results: the expression of bFGF was weakly positive in control group and 鹵2Gz exposed 30min group. The expression of bFGF in hippocampal neurons of rats exposed to 鹵6Gz and 鹵8Gz showed that the expression of bFGF began to increase at 6 h after exposure, and the expression of bFGF in experimental group decreased significantly after 24 h of exposure, but it was still higher than that in normal control group. Compared with the control group, the difference was significant (P 0. 05). Conclusion: the expression of bFGF in rat hippocampal neurons can be increased by high-intensity push-pull exercise.
