核因子κB在肺型氧中毒發(fā)病中的作用研究

【摘要】:在潛水高氣壓作業(yè)及臨床高壓氧(hyperbaric oxygen,HBO)治療中,都涉及機體在高分壓氧中暴露的問題。高分壓氧的應用可以提高減壓時惰性氣體脫飽和速率,縮短減壓時間,提高水下作業(yè)效率,并且對一些缺氧性疾病有明確的治療效果,但是長時間吸入高分壓氧又帶來氧中毒的問題。氧中毒是機體吸入高于50kPa的氧氣一定時間后,導致機體某些器官的結(jié)構(gòu)與功能發(fā)生病理性變化而表現(xiàn)的病癥。氧中毒分為眼型、腦型、和肺型三種類型,其中肺型氧中毒的主要表現(xiàn)為咳嗽、胸骨后疼痛、呼吸困難,形態(tài)學觀察發(fā)現(xiàn)早期以炎癥滲出為主,表現(xiàn)為肺部出現(xiàn)大面積充血、出血和水腫,炎細胞浸潤。對于氧中毒的發(fā)病機理,至今仍未完全明了。 核因子κB(NF-κB)是一個多功能核轉(zhuǎn)錄因子,具有廣泛的生物學活性,激活后可促進細胞因子、粘附分子、趨化因子等基因轉(zhuǎn)錄,在應激、炎癥和免疫反應等過程中發(fā)揮重要作用,成為近年來研究的熱點。最近研究發(fā)現(xiàn)高分壓氧能夠?qū)е翹F-κB活化。 腫瘤壞死因子-α(tumor necrosis factor-alpha,TNF-α)是受NF-κB調(diào)控表達的具有炎癥介導活性的一種細胞因子,TNF-α的高表達可以造成肺損傷。 本實驗目的是通過研究NF-κB和TNF-α在氧中毒時的改變規(guī)律,確定NF-κB活化在肺型氧中毒發(fā)病中的作用,探討肺型氧中毒的可能機制,為預防肺型氧中毒提供新的思路。 本研究采用健康SD大鼠在密閉氧艙中吸入高壓氧的方法建立肺型氧中毒模型。實驗共分6組,分別是正常對照組、常氧高氮對照組、0.23MPa高壓氧暴露2h、6h、
[Abstract]:In the treatment of high pressure diving and hyperbaric oxygen (HBO) treatment, it is involved in the exposure of the body to high partial pressure oxygen (HBO). The application of high partial pressure oxygen can improve the desaturation rate of inert gas during decompression, shorten the decompression time, improve the efficiency of underwater operation, and have definite therapeutic effect on some hypoxic diseases. But long-term inhalation of high partial pressure oxygen also leads to oxygen poisoning. Oxygen poisoning is a kind of disease caused by pathological changes in the structure and function of some organs after inhaling oxygen above 50 KPA for a certain time. Oxygen poisoning can be divided into three types: eye type, brain type, and lung type. The main manifestations of oxygen poisoning in pulmonary type are cough, pain after sternum, dyspnea. Bleeding and edema, inflammatory cell infiltration. The pathogenesis of oxygen poisoning has not been fully understood. NF-魏 B is a multifunctional nuclear transcription factor with a wide range of biological activities. Activation can promote transcription of cytokines, adhesion molecules, chemokines and other genes, and play an important role in stress, inflammation and immune response. It has become a hot topic in recent years. Recent studies have found that high partial pressure oxygen can lead to NF- 魏 B activation. Tumor necrosis factor-alpha (TNF- 偽) is a cytokine with inflammatory mediated activity regulated by NF- 魏 B, which can cause lung injury. The purpose of this study was to study the changes of NF- 魏 B and TNF- 偽 in oxygen poisoning, to determine the role of NF- 魏 B activation in the pathogenesis of pulmonary oxygen poisoning, to explore the possible mechanism of pulmonary oxygen poisoning, and to provide a new way of thinking for the prevention of pulmonary oxygen poisoning. A model of pulmonary oxygen poisoning was established by inhaling hyperbaric oxygen in a closed oxygen chamber in healthy SD rats. The experiment was divided into 6 groups: normal control group and normoxic hyperbaric oxygen control group with 0.23 MPA hyperbaric oxygen exposure for 2 h or 6 h, respectively.
