鈣蛋白酶介導(dǎo)模擬失重大鼠心肌肌鈣蛋白抑制亞基的降解

【摘要】:本文旨在觀察尾部懸吊模擬失重大鼠心肌鈣蛋白酶(calpain)與鈣蛋白酶抑素(calpastatin)表達(dá)的變化,以探討心肌肌鈣蛋白抑制亞基(cardiac troponin I,cTnI)降解的可能機(jī)制。采用尾部懸吊模擬失重大鼠模型,Western blotting技術(shù)觀測(cè)心肌calpain-1、calpain-2與calpastatin的表達(dá);PD150606抑制calpain活性,分析cTnI降解程度的變化。結(jié)果顯示:與同步對(duì)照組相比,懸吊2周與4周組大鼠心肌calpastatin表達(dá)呈顯著性降低(P0.05),calpain-1表達(dá)未改變,calpain-2表達(dá)略有降低;但是,心肌calpain-1/calpastatin及calpain-2/calpastatin的比值在懸吊2周與4周組明顯增高(P0.05,P0.01)。懸吊4周組cTnI降解顯著高于對(duì)照組(P0.01);然而,用calpain非特異性抑制劑PD150606處理后,對(duì)照組及懸吊組cTnI的降解均被顯著抑制(P0.01)。這些結(jié)果提示模擬失重大鼠心肌calpain活性增高可能增加cTnI的降解。
[Abstract]:The aim of this study was to investigate the changes of cardiac calpainin and calpastatin expression in tail suspended simulated weightlessness rats, in order to explore the possible mechanism of cardiac troponin inhibiting the degradation of cardiac troponin IcTnI. Western blotting technique was used to observe the expression of calpain-1 calpain-2 and calpastatin in rat model of simulated weightlessness by tail suspension. PD150606 inhibited the activity of calpain and analyzed the degradation degree of cTnI. The results showed that compared with the control group, the expression of calpastatin in rat myocardium was significantly decreased after 2 and 4 weeks of suspension, but the ratio of calpain-1/calpastatin to calpain-2/calpastatin was significantly increased in the groups of 2 weeks and 4 weeks of suspension, but the expression of calpain-2 was not changed slightly, however, the ratio of calpain-1/calpastatin to calpain-2/calpastatin was significantly increased in the groups of 2 weeks and 4 weeks of suspension. The degradation of cTnI in the suspension group was significantly higher than that in the control group (P 0.01), however, the degradation of cTnI in both the control group and the suspension group was significantly inhibited after treatment with PD150606, a non-specific inhibitor of calpain. These results suggest that the increase of myocardial calpain activity may increase the degradation of cTnI in simulated weightlessness rats.
