中藥干預模擬失重大鼠股骨外源鈣沉積研究

【摘要】:目的:研究中藥復方對模擬失重大鼠承重骨對外源鈣攝取的干預作用。方法:雄性Wistar大鼠隨機分為3組,空白組、模型組、中藥組。尾吊模擬失重,中藥復方(含熟地黃、懷牛膝、黃芪、當歸、牡蠣醋酸水解物等)704 g.L-1水煎劑2.5mL灌胃;適應期1周,模擬失重期3周,于模擬失重第11天一次性灌胃給予一定量41Ca示蹤劑。實驗結束取股骨,加速器質譜法(Accelerator Mass Spectrometry,AMS)分析骨41Ca/40Ca值,電感耦合等離子體光譜法(Inductively Coupled Plasma-AtomicEmission Spectroscopy,ICP-AES)分析骨鈣總量,計算股骨41Ca沉積量、股骨41Ca沉積率。結果:空白、模型、中藥3組股骨41Ca/40Ca豐度比分別為(5.03±0.55),(3.82±0.29),(5.73±0.40)×10-9,模型組較空白組顯著降低(P0.001),中藥組較空白組顯著升高(P0.05);空白、模型、中藥3組股骨41Ca沉積量分別為(7.39±0.90),(4.52±0.40),(7.45±0.89)×10-7mg,沉積率分別為(0.819±0.100),(0.501±0.045),(0.826±0.098)%,模型組股骨41Ca沉積量和沉積率均較空白組顯著降低(P0.001),中藥組較空白組變化不顯著,較模型組顯著增高(P0.001)。結論:尾吊模擬失重可造成大鼠承重骨外源鈣沉積障礙,中藥可較好對抗這種情況;失重骨質疏松可能存在外源鈣沉積減少以外的多種機制。
[Abstract]:Aim: to study the effect of Chinese medicine compound on exogenous calcium uptake in weight-bearing bone of simulated weightlessness rats.Methods: male Wistar rats were randomly divided into 3 groups: blank group, model group and traditional Chinese medicine group.Tail suspension simulated weightlessness, traditional Chinese medicine compound (including Radix Rehmanniae, Achyranthes bidentata, Radix Astragali, Angelica sinensis, oyster acetic acid hydrolysate, etc.) 2.5mL, the adaptation period was 1 week, and the simulated weightlessness period was 3 weeks.A certain amount of 41Ca tracer was given once on the 11th day of simulated weightlessness.At the end of the experiment, the femur was analyzed by Accelerator Mass Spectrometry (AMS), and the total bone calcium was analyzed by inductively Coupled Plasma-AtomicEmission spectroscopy (ICP-AESS-MS). The deposition of 41Ca in femur and 41Ca deposition rate in femur were calculated.Results: the 41Ca/40Ca abundance ratio of femur in the blank, model and TCM groups was 5.03 鹵0.55, 3.82 鹵0.29, 5.73 鹵0.40) 脳 10 ~ (-9), respectively. Compared with the blank group, the 41Ca/40Ca abundance ratio of the model group was significantly lower than that of the blank group, and that of the traditional Chinese medicine group was significantly higher than that of the blank group, while the blank, the model group was significantly higher than that of the blank group.The deposition amount of 41Ca in the femur of the three groups were 7.39 鹵0.90, 4.52 鹵0.40, 7.45 鹵0.89) 脳 10 ~ (-7) mg, and the deposition rates were 0.819 鹵0.100 鹵0.501 鹵0.045, 0.826 鹵0.098, respectively. The 41Ca deposition and deposition rate of the femur in the model group were significantly lower than those in the blank group (P 0.001), but the changes in the traditional Chinese medicine group were not significant compared with the blank group, and significantly higher than that in the model group.Conclusion: tail suspension simulated weightlessness can cause exogenous calcium deposition obstacle in load-bearing bone of rats, Chinese medicine can better resist this situation, and weightlessness osteoporosis may have some mechanisms other than the reduction of exogenous calcium deposition.
