循環(huán)血miR-21在空間輻射監(jiān)測中的應(yīng)用潛力

【摘要】:深空探測中如何在輻射暴露早期用快速、簡便的方法進(jìn)行輻射損傷及風(fēng)險評估問題,對傳統(tǒng)的輻射劑量計(jì)提出巨大的挑戰(zhàn)。循環(huán)血microRNA(miRNA)具有很好的特異性和穩(wěn)定性,且積極響應(yīng)外界環(huán)境變化,因此,篩選循環(huán)血中輻射敏感的miRNA,對開發(fā)基于血液的微創(chuàng)生物輻射標(biāo)志物具有較大的研究意義和應(yīng)用價值。本文利用X射線對昆明小鼠進(jìn)行全身照射,24小時后取血,利用miRNA PCR array的方法檢測了21種候選miRNA的表達(dá)譜水平,發(fā)現(xiàn)多種miRNA對輻射有響應(yīng),其中miR-21具有較大的變化幅度。利用實(shí)時定量PCR分別驗(yàn)證了miR-21在不同射線(X射線、碳離子束、鐵離子束)條件下的劑量效應(yīng)和時間效應(yīng)。檢測了miR-21在相同和不同鼠齡小鼠個體間的本底表達(dá)水平。結(jié)果表明,miR-21在不同類型射線照射下表達(dá)水平都有升高的趨勢,特別是對低劑量輻射也具有顯著的響應(yīng),具有較強(qiáng)的劑量效應(yīng)關(guān)系。在6~72小時內(nèi),其表達(dá)水平維持在一個穩(wěn)定的閾值內(nèi)。miR-21在相同和不同鼠齡小鼠個體間的本底表達(dá)水平不具有差異性,在血清樣本的保存及RNA的提取過程中具有較強(qiáng)的穩(wěn)定性。以上特征說明miR-21具有作為生物輻射劑量計(jì)并應(yīng)用于輻射損傷檢測的潛力。
[Abstract]:In deep space exploration, the rapid and simple method of radiation damage and risk assessment in the early stage of radiation exposure poses a great challenge to the traditional radiation dose measurement. Circulating blood microRNA (miRNA) has good specificity, stability and positive response to environmental changes. Therefore, screening radiosensitive miRNA, in circulating blood has great significance and application value in the development of microinvasive biomarkers based on blood. In this paper, blood samples were taken from Kunming mice after whole-body irradiation by X-ray for 24 hours. The expression levels of 21 candidate miRNA were detected by miRNA PCR array method. It was found that many kinds of miRNA were responsive to radiation, among which miR-21 had a large range of changes. The dose effect and time effect of miR-21 under different radiation conditions (X-ray, carbon ion beam, iron ion beam) were verified by real-time quantitative PCR. The background expression of miR-21 in mice of the same and different ages was detected. The results showed that the expression level of miR-21 increased under different types of radiation, especially in response to low dose radiation, and had a strong dose-effect relationship. The expression level of miR-21 was maintained within a stable threshold within 6h for 72 hours. The expression level of miR-21 was not different in the same and different mouse aged mice, but had strong stability in the preservation of serum samples and the process of RNA extraction. These characteristics indicate that miR-21 has the potential to be used as a biological dosimeter and to be used in radiation damage detection.
