不同載荷強(qiáng)度對(duì)骨腱結(jié)合部結(jié)構(gòu)的影響

【摘要】:研究目的:目前運(yùn)動(dòng)受到越來(lái)越廣泛的重視,更多的人參與到運(yùn)動(dòng)中,而競(jìng)技運(yùn)動(dòng)的水平也在日益增高,同時(shí)不可避免的出現(xiàn)各種跟運(yùn)動(dòng)相關(guān)的病癥,骨腱結(jié)合部的損傷是困擾運(yùn)動(dòng)訓(xùn)練和人們?nèi)粘I畹某R?jiàn)損傷,也是體育科研領(lǐng)域的難題,本論文通過(guò)不同強(qiáng)度的循環(huán)載荷刺激作為干預(yù)手段,對(duì)骨腱結(jié)合部損傷進(jìn)行研究,確定不會(huì)引起骨腱結(jié)合部損傷的刺激強(qiáng)度閡值。 研究對(duì)象與方法:本研究的實(shí)驗(yàn)對(duì)象為40只成年(18周)雌性新西蘭大白兔,體重2.5±0.2kg,隨機(jī)篩選分為循環(huán)載荷CL30(n=8), CL24(n=8), CL18(n=8), CL12(n=8), CL6(n=8),5個(gè)強(qiáng)度組,左側(cè)后肢為循環(huán)載荷組,右側(cè)后肢為空白對(duì)照組CON (n=8),通過(guò)循環(huán)載荷損傷造模的方法,每次訓(xùn)練刺激2小時(shí)(收縮7200次),每周3次,共4周,累積24小時(shí),4周訓(xùn)練結(jié)束后,動(dòng)物處死取材,脫鈣,脫水,石蠟包埋,切片,通過(guò)HE, Safranin0組織學(xué)染色以及VEGF、 bFGF免疫組化染色的方法對(duì)髕腱骨腱結(jié)合部的組織學(xué)變化進(jìn)行觀察,并且利用圖像處理軟件對(duì)所采集的圖像進(jìn)行分析。評(píng)價(jià)指標(biāo)有:肌腱細(xì)胞密度、纖維軟骨帶厚度、粘多糖蛋白分布以及VEGF、bFGF免疫組化染色光密度值。 結(jié)果:4周的循環(huán)載荷刺激后,HE染色結(jié)果顯示,載荷組CL30, CL24, CL12,CL6組與CON組比較,細(xì)胞密度顯著性增高,CL18組與對(duì)照組相比沒(méi)有顯著性差異;CL30和CL24組纖維軟骨帶厚度與CON組比較明顯增厚,有顯著性差異。VEGF免疫組化染色:載荷組CL30組VEGF的表達(dá)與CON比較顯著性升高,CL24、CL18、CL12、CL6與CON組比較沒(méi)有顯著性差異。bFGF免疫組化染色:循環(huán)載荷組CL12、CL6組bFGF的表達(dá)與CON組比較顯著性增高。 結(jié)論:1.通過(guò)4周不同強(qiáng)度的循環(huán)載荷刺激,CL3O、CL24組出現(xiàn)了損傷的病理變化,隨著運(yùn)動(dòng)強(qiáng)度的不同,骨腱結(jié)合部發(fā)生了相應(yīng)的結(jié)構(gòu)改變,說(shuō)明骨腱結(jié)合部損傷與運(yùn)動(dòng)強(qiáng)度相關(guān)。2.CL12組是骨腱結(jié)合部損傷發(fā)生的閡下強(qiáng)度,CL12、CL6組的循環(huán)載荷刺激都不會(huì)造成骨腱結(jié)合部損傷,以此為以后運(yùn)動(dòng)訓(xùn)練和健身的損傷預(yù)防提供依據(jù)。
[Abstract]:Research objective: at present, more and more people pay attention to sports, more people participate in sports, and the level of competitive sports is increasing day by day. At the same time, there are inevitably various diseases related to sports. The injury of the bone tendon joint is a common injury which puzzles the exercise training and people's daily life, and is also a difficult problem in the field of sports scientific research. In this paper, the injury of the bone tendon joint is studied by the different intensity cyclic load stimulation as the intervention method. Determine the threshold of stimulation intensity that does not cause injury to the bone tendon junction. Subjects and methods: 40 adult (18 weeks) female New Zealand white rabbits, weighing 2.5 鹵0.2 kg, were randomly divided into four groups: cyclic load CL30 (NN8), CL24 (NC8), CL18 (NC8), CL12 (NC8), CL6 (NN8), 5 intensity groups, and left hind limb as cyclic loading group. The right hind limb was a blank control group (CON 8). By the method of cyclic load injury, the rats were stimulated for 2 hours (contraction 7200 times), 3 times a week for a total of 4 weeks. After 24 hours and 4 weeks of training, the animals were sacrificed, decalcified and dehydrated. The histological changes of the patellar tendon joint were observed by HE, Safranin0 histological staining and VEGF, bFGF immunohistochemical staining, and the images were analyzed by image processing software. The indexes were as follows: the density of tendon cells, the thickness of fibrochondrocytes, the distribution of mucopolysaccharide protein and the optical density of VEGF,bFGF immunohistochemical staining. Results the cell density of CL30, CL24, CL12,CL6 group was significantly higher than that of CON group. There was no significant difference in cell density between CL18 group and control group. The thickness of fibrochondrocytes in CL30 and CL24 groups was significantly thicker than that in CON group. The expression of VEGF in CL30 group was significantly higher than that in CON group. The expression of bFGF in CL12,CL6 group was significantly higher than that in CON group. Conclusion 1. The pathological changes of injury were observed in the CL3OOG CL24 group after 4 weeks of cyclic load stimulation. With the different intensity of exercise, the corresponding structural changes occurred in the bony tendon junctions. The results showed that the injury of bone tendon joint was related to the intensity of exercise. 2. Group CL12 was the group of bone tendon joint injury. The cyclic load stimulation of group CL12 and CL6 would not cause the injury of bone tendon junction, which would provide the basis for the prevention of injury in later exercise training and fitness.
