加味酸棗仁湯對(duì)運(yùn)動(dòng)性失眠大鼠單胺類神經(jīng)遞質(zhì)影響的研究

【摘要】:1.研究目的 通過觀察運(yùn)動(dòng)性失眠大鼠服用復(fù)方中藥后其整體特征及腦組織單胺類神經(jīng)遞質(zhì)的動(dòng)態(tài)變化,探討復(fù)方中藥對(duì)單胺類神經(jīng)遞質(zhì)代謝的影響,更好的認(rèn)識(shí)單胺類神經(jīng)遞質(zhì)在睡眠-覺醒節(jié)律調(diào)節(jié)中的作用以及運(yùn)動(dòng)性失眠產(chǎn)生的機(jī)制,為有效解決運(yùn)動(dòng)性失眠和指導(dǎo)運(yùn)動(dòng)訓(xùn)練提供科學(xué)依據(jù)。 2.研究方法 選取8周齡健康雌性wistar大鼠60只,隨機(jī)分為對(duì)照組、運(yùn)動(dòng)性失眠模型組、自然康復(fù)組、加味酸棗仁湯低劑量組、加味酸棗仁湯高劑量組、安定片組,每組10只。6周相關(guān)干預(yù)后各組大鼠斷頭處死,開顱取腦。采用高效液相色譜儀-熒光檢測(cè)器檢測(cè)大鼠腦組織內(nèi)5-羥色胺、5-羥吲哚乙酸、多巴胺、去甲腎上腺素的含量。所有數(shù)據(jù)采用SPSS17.0統(tǒng)計(jì)軟件進(jìn)行統(tǒng)計(jì)學(xué)處理。 3.實(shí)驗(yàn)結(jié)果 3.1與對(duì)照組比較,模型組大鼠腦組織內(nèi)單胺類遞質(zhì)5-HT及其代謝產(chǎn)物5-HIAA含量均極明顯降低(p0.001),NE和DA的含量及DA/5-HT的值則明顯升高(p0.01或p0.001)。 3.2與模型組比較,加味酸棗仁湯高劑量組大鼠腦組織內(nèi)5-HT及其代謝產(chǎn)物5-HIAA含量均極明顯升高(p0.001),NE和DA的含量及DA/5-HT的值則極明顯降低(p0.001)。 3.3與西藥組比較,加味酸棗仁湯高劑量組大鼠腦組織內(nèi)5-HT及其代謝產(chǎn)物5-HIAA含量明顯升高(p0.05),,NE含量及DA/5-HT的值則明顯降低(p0.05)。 3.4與對(duì)照組比較,在建模后期,其他各組實(shí)驗(yàn)大鼠體重與攝食量均顯著性降低(p0.001),經(jīng)加味酸棗仁湯治療后,雖都有所增加,但還是非常明顯低于對(duì)照組(p0.01)。 4.結(jié)論 4.1采用大負(fù)荷游泳訓(xùn)練+腹腔注射PCPA所致的失眠大鼠模型,符合中醫(yī)學(xué)虛勞失眠癥候模型,表明采用該方法建立動(dòng)物運(yùn)動(dòng)失眠模型具有較強(qiáng)的可行性。 4.2運(yùn)動(dòng)性失眠可導(dǎo)致大鼠體重和攝食量的減少,加味酸棗仁湯能促進(jìn)大鼠運(yùn)動(dòng)疲勞的恢復(fù)和改善大鼠睡眠和攝食質(zhì)量。 4.3運(yùn)動(dòng)性失眠可以導(dǎo)致大鼠腦組織內(nèi)單胺類神經(jīng)遞質(zhì)5-HT及其代謝產(chǎn)物5-HIAA含量明顯降低,而NE、DA及DA/5-HT的值明顯增加。加味酸棗仁湯干預(yù)可提高腦組織5-HT和5-HIAA含量,降低NE、DA及DA/5-HT的值,改善睡眠,保護(hù)大腦。而且加味酸棗仁湯的療效與量效關(guān)系密切。
[Abstract]:1. Objective to investigate the effects of compound Chinese medicine on the metabolism of monoamine neurotransmitters by observing the overall characteristics of rats with exercise insomnia and the dynamic changes of monoamine neurotransmitters in brain tissue. To better understand the role of monoamine neurotransmitters in the regulation of sleep-arousal rhythm and the mechanism of motor insomnia. 2. Methods Sixty healthy female wistar rats aged 8 weeks were randomly divided into three groups: control group, model group of exercise insomnia, natural rehabilitation group, low dose group of modified Suanzaoren decoction, high dose group of modified Suanzaoren decoction and diazepam tablet group. 10 rats in each group were killed after 6. 6 weeks of intervention and craniotomy. The contents of 5-hydroxyindoleacetic acid, dopamine and norepinephrine in rat brain were determined by high performance liquid chromatograph and fluorescence detector. All the data were processed by SPSS 17.0 statistical software. Results 3.1 compared with the control group, the contents of monoamine transmitter 5-HT and its metabolite 5-HIAA in the brain of the model group were significantly decreased (p0.001), the contents of NE and DA, and the value of DA-5-HT were significantly increased (p0.01 or p0.001). 3.2 compared with the model group, the contents of 5-HT and 5-HIAA in the brain tissue of the model group were significantly higher than those in the model group. The contents of 5-HT and its metabolite 5-HIAA in brain tissue of rats with high dosage of Jiawei Suanzaoren decoction were significantly increased (p0.001), the contents of NE and DA, and the value of DA-5-HT were significantly decreased (p0.001). 3.3 compared with western medicine group, the content of 5-HT and 5-HIAA in brain tissue of rats were significantly lower than those in western medicine group. The contents of 5-HT and its metabolite 5-HIAA in brain tissue of rats with high dosage of Jiawei Suanzaoren decoction were significantly increased (p0.05) and the value of DA-5-HT were significantly decreased (p0.05). 3.4 compared with the control group, the content of 5-HT and its metabolite 5-HIAA were significantly decreased in the later stage of modeling. The body weight and food intake of other experimental rats were significantly decreased (p0.001). After treated with modified Suanzaoren decoction, the rats were all increased, but still significantly lower than the control group (p0.01). Conclusion 4.1 Insomnia rat model induced by intraperitoneal injection of PCPA with heavy load swimming training is in line with the model of asthenia insomnia in traditional Chinese medicine. The results showed that it was feasible to establish the animal model of exercise insomnia by using this method. 4.2 exercise insomnia could reduce the body weight and food intake of rats. Jiawei Suanzaoren decoction could promote the recovery of exercise fatigue and improve the sleep and feeding quality of rats. 4.3 the content of monoamine neurotransmitter 5-HT and its metabolite 5-HIAA in rat brain was significantly decreased after exercise insomnia. However, the values of DA and DA / D / 5 HT increased significantly. The intervention of Jiawei Suanzaoren decoction could increase the contents of 5-HT and 5-HIAA in brain tissue, decrease the values of NEDA and DA-5-HT, improve sleep and protect the brain. Moreover, the curative effect of modified Suanzaoren decoction is closely related to its dose effect.
