補(bǔ)充活性肽對(duì)骨骼肌Calpain基因表達(dá)的影響

【摘要】:1研究目的本實(shí)驗(yàn)通過觀察補(bǔ)充活性肽對(duì)Calpain-1.Calpain-2基因表達(dá)的影響來探討活性肽在預(yù)防骨骼肌蛋白降解中所發(fā)揮的作用,另外還期望在亞細(xì)胞水平對(duì)骨骼肌微損傷的生理調(diào)節(jié)機(jī)制進(jìn)行一定程度的闡釋 2研究方法8周齡雄性SD大鼠隨機(jī)分為安靜對(duì)照組,運(yùn)動(dòng)后即刻、24h、48h對(duì)照組,運(yùn)動(dòng)后即刻、24h、48h+海洋肽實(shí)驗(yàn)組,運(yùn)動(dòng)后即刻、24h、48h+大豆蛋白陽性對(duì)照組,共10組,每組10只,進(jìn)行6周跑臺(tái)訓(xùn)練。采用放射免疫法測定血清T.IGF-1含量;采用Real Time-PCR法測定骨骼肌Calpain-1.Calpain-2mRNA含量。 3研究結(jié)果3.1運(yùn)動(dòng)后,海洋肽運(yùn)動(dòng)組血清IGF-1與單純運(yùn)動(dòng)組、大豆蛋白運(yùn)動(dòng)組無顯著性差異;隨時(shí)間變化,運(yùn)動(dòng)后即刻與運(yùn)動(dòng)前差異顯著(P0.05)。3.2運(yùn)動(dòng)后24h,海洋肽運(yùn)動(dòng)組血清睪酮與單純運(yùn)動(dòng)組差異顯著(P0.05);隨時(shí)間變化無顯著性差異。3.3海洋肽運(yùn)動(dòng)組運(yùn)動(dòng)后即刻股中肌CANP-1基因表達(dá)變化與單純運(yùn)動(dòng)組、大豆蛋白運(yùn)動(dòng)組差異顯著(P0.05);隨時(shí)間變化,差異非常顯著(P0.05)。運(yùn)動(dòng)后24h,股中肌CANP-2基因表達(dá)變化與大豆蛋白運(yùn)動(dòng)組差異顯著(P0.05);隨時(shí)間變化,無顯著性差異。3.4海洋肽運(yùn)動(dòng)組運(yùn)動(dòng)后24h,腓腸肌CANP-1基因表達(dá)變化與單純運(yùn)動(dòng)組差異顯著(P0.05);運(yùn)動(dòng)后48h,與大豆蛋白運(yùn)動(dòng)組差異顯著(P0.01);隨時(shí)間變化,無顯著性差異。運(yùn)動(dòng)后即刻、運(yùn)動(dòng)后48h,腓腸肌CANP-2基因表達(dá)變化與大豆蛋白運(yùn)動(dòng)組差異顯著(P0.01);隨時(shí)間變化,差異非常顯著(P0.01)。 4研究結(jié)論4.1海洋肽與大豆蛋白均能夠縮短運(yùn)動(dòng)后血清IGF-1、血清T的恢復(fù)時(shí)間;海洋肽的效果好于大豆蛋白,促進(jìn)睪酮恢復(fù)的速率更快。4.2運(yùn)動(dòng)后,骨骼肌CANP-1、CANP-2基因表達(dá)呈現(xiàn)不同變化;CANP-1基因表達(dá)隨時(shí)間延長持續(xù)下調(diào),而CANP-2基因表達(dá)上調(diào)。4.3海洋肽在運(yùn)動(dòng)過程中對(duì)骨骼肌有一定的保護(hù)作用,而大豆蛋白主要在運(yùn)動(dòng)后48h的炎癥反應(yīng)階段發(fā)揮保護(hù)作用。4.4運(yùn)動(dòng)后股中肌CANP-1基因表達(dá)持續(xù)下調(diào),而腓腸肌CANP-1.CANP-2基因表達(dá)呈現(xiàn)為先增高后降低,這與肌纖維類型有關(guān)。
[Abstract]:Objective to investigate the role of active peptides in the prevention of skeletal muscle protein degradation by observing the effect of supplementation of active peptides on the expression of Calpain-1.Calpain-2 gene. In addition, it is expected to explain to a certain extent the physiological regulation mechanism of skeletal muscle microdamage at the subcellular level. Methods the 8-week-old male SD rats were randomly divided into quiet control group and control group for 24 h and 48 h immediately after exercise. Immediately after exercise, 10 rats in 10 groups were treated with marine peptide for 24 hours and 48 hours after exercise, and the control group with soybean protein positive for 24 hours and 48 hours after exercise. 10 rats in each group were trained by running platform for 6 weeks. The levels of serum T.IGF-1 and Calpain-1.Calpain-2 mRNA in skeletal muscle were measured by radioimmunoassay and Real Time-PCR respectively. 3 the results showed that there was no significant difference in serum IGF-1 between marine peptide exercise group and soybean protein exercise group after 3.1 exercise. There were significant differences in serum testosterone between exercise group and exercise group (P0.05) immediately after exercise and 24 hours after exercise (P0.05). There was no significant difference with time. 3.3 the changes of CANP-1 gene expression in middle femoral muscle immediately after exercise in marine peptide exercise group were significantly different from those in simple exercise group and soybean protein exercise group (P0.05), and the difference was very significant with time (P0.05). The changes of CANP-2 gene expression in middle femoris muscle were significantly different from those in soybean protein exercise group at 24 h after exercise (P0.05), but there was no significant difference between exercise group and exercise group at 24 h after exercise (P0.05), and there was no significant difference in CANP-1 gene expression in gastrocnemius muscle between exercise group and exercise group 24 hours after exercise (P0.05). There was a significant difference between the exercise group and the soybean protein group 48 hours after exercise (P0.01), but there was no significant difference with time. The changes of CANP-2 gene expression in gastrocnemius muscle immediately after exercise and 48 hours after exercise were significantly different from those in soybean protein exercise group (P0.01). The difference was very significant (P0.01). 4 conclusion 4.1 Marine peptide and soybean protein can shorten the recovery time of serum IGF-1 and serum T after exercise, the effect of marine peptide is better than that of soybean protein, and the recovery rate of testosterone is faster after exercise .4.2. The expression of CANP-1 and CANP-2 in skeletal muscle showed different changes. CANP-1 gene expression continued to be down-regulated over time, while CANP-2 gene expression up-regulated .4.3 marine peptide had protective effect on skeletal muscle during exercise. The expression of CANP-1 gene in femur muscle decreased continuously after exercise 48 h after exercise, while CANP-1. CANP-2 gene expression in gastrocnemius muscle increased first and then decreased, which was related to the type of muscle fiber.
