低功率激光對(duì)一次性離心力竭運(yùn)動(dòng)大鼠跟腱微損傷的修復(fù)作用

【摘要】:研究目的:急性跟腱損傷是一直困擾運(yùn)動(dòng)員的一個(gè)棘手問題。一次性離心力竭運(yùn)動(dòng)跟腱微損傷作為急性跟腱損傷的代表,關(guān)于其修復(fù)機(jī)制研究還不是很清楚。本研究采用低功率激光對(duì)一次性離心力竭運(yùn)動(dòng)大鼠跟腱進(jìn)行照射,采用生物化學(xué)測(cè)定和組織學(xué)技術(shù)觀察的方法,分析低功率激光照射對(duì)跟腱微損傷修復(fù)變化時(shí)的特點(diǎn),探討低功率激光照射對(duì)力竭運(yùn)動(dòng)后早期跟腱微損傷修復(fù)過程中的作用。 研究方法:60只8周齡雄性Wistar大鼠,隨機(jī)分為兩組,分別為空白對(duì)照組(n=6)和一次性離心力竭跑臺(tái)運(yùn)動(dòng)組(n=54),其中一次性離心力竭跑臺(tái)運(yùn)動(dòng)組分為運(yùn)動(dòng)后即刻組(n=6),自然愈合1天組(n=6)、2天組(n=6)、3天組(n=6)、7天組(n=6),激光治療1天組(n=6)、2天組(n=6)、3天組(n=6)、7天組(n=6)。力竭跑臺(tái)運(yùn)動(dòng)后于第1天,第2天、第3天、第7天分別固定時(shí)間進(jìn)行一次激光照射。采用堿水解法測(cè)定各組跟腱羥脯氨酸含量,采用ELISA法測(cè)定各組跟腱Ⅰ型膠原、Ⅲ型膠原以及IGF-Ⅰ蛋白含量,采用HE染色法通過光鏡觀察跟腱中膠原纖維狀態(tài)及腱細(xì)胞有無炎性變化,通過透射電鏡觀察膠原原纖維排列、腱細(xì)胞、細(xì)胞核及細(xì)胞器等變化情況。 研究結(jié)果:(1)光鏡和透射電鏡觀察結(jié)果表明低功率激光能夠提高腱細(xì)胞分泌功能、對(duì)跟腱微損傷的修復(fù)具有積極作用;(2)一次性離心力竭運(yùn)動(dòng)大鼠跟腱造模對(duì)照組羥脯氨酸、Ⅰ型膠原、Ⅲ型膠原含量與空白對(duì)照組相比均有顯著性差異(P0.05),其結(jié)果表明造模成功;(3)激光治療3天組跟腱羥脯氨酸含量顯著高于自然愈合3天組(P0.05),激光治療7天組顯著低于自然愈合7天組(P0.05),而激光治療1、2天組與自然愈合1、2組無明顯差異(P0.05);(4)激光治療1、2、3天組跟腱IGF-Ⅰ含量均分別顯著高于自然愈合1、2、3天組(P0.05);激光治療7天組顯著低于自然愈合7天組(P0.05);(5)激光治療組Ⅰ型膠原含量變化趨勢(shì)與自然愈合組相同,但激光治療3、7天組顯著高于自然愈合3、7天組(P0.05)而激光治療組Ⅲ型膠原含量于2天時(shí)達(dá)到峰值,顯著高于自然愈合組(P0.05),激光治療3、7天組Ⅲ型膠原均顯著低于自然愈合3、7組(P0.05)。 研究結(jié)論:低功率激光照射對(duì)一次性離心力竭運(yùn)動(dòng)大鼠跟腱微損傷修復(fù)具有一定促進(jìn)作用;低功率激光照射能夠提高一次性離心力竭運(yùn)動(dòng)大鼠跟腱膠原合成,,調(diào)節(jié)細(xì)胞外膠原代謝,加快跟腱微損傷的修復(fù)。
[Abstract]:Objective: acute Achilles tendon injury has been a thorny problem for athletes. As the representative of acute Achilles tendon injury, the study of repair mechanism is not clear. In this study, low power laser was used to irradiate the Achilles tendon of rats during one time centrifugal exhaustive exercise. The characteristics of microdamage repair of Achilles tendon by low power laser irradiation were analyzed by means of biochemical measurement and histological observation. To investigate the effect of low power laser irradiation on the repair of early microinjury of Achilles tendon after exhaustive exercise. Methods Sixty 8-week-old male Wistar rats were randomly divided into two groups. They were blank control group (NX6) and once centrifuge exhaustive treadmill exercise group (NN6). Among them, once centrifuge exhausted treadmill exercise group was divided into three groups: immediate after exercise group (NQ6), natural healing day group (NN6), 3 day group (NN6) and 7 day group (N6), laser treatment group for 1 day. (NJ 6) 2 days group (ng 6) 3 days group (ng 6) and 7 d group (ng 6).
