S-異戊烯基-L-半胱氨酸對輻射誘導(dǎo)小鼠DNA損傷的保護(hù)作用

【摘要】:目的采用動物急性輻射損傷模型評估S-異戊烯基-L-半胱氨酸對輻射誘導(dǎo)DNA損傷的保護(hù)作用。方法將40只ICR小鼠隨機(jī)分為空白對照組、1.0Gy伽馬射線照射組、1.0Gy伽馬射線照射聯(lián)合S-異戊烯基-L-半胱氨酸給藥組、7.2Gy伽馬射線照射組、7.2Gy伽馬射線照射聯(lián)合S-異戊烯基-L-半胱氨酸給藥組5組,每組8只,采用外周血淋巴細(xì)胞彗星實(shí)驗(yàn)和骨髓嗜多染紅細(xì)胞微核實(shí)驗(yàn)檢測1.0Gy和7.2Gy伽馬射線照射后不同處理組小鼠DNA雙鏈斷裂情況。結(jié)果7.2Gy伽馬射線照射組小鼠外周血淋巴細(xì)胞的尾部DNA百分含量、尾長、尾矩和Olive尾矩明顯高于空白對照組(P均0.01)。7.2Gy伽馬射線照射聯(lián)合S-異戊烯基-L-半胱氨酸給藥組的DNA百分含量、尾長、尾矩和Olive尾矩明顯低于7.2Gy伽馬射線照射組(P均0.05)。1.0Gy伽馬射線照射組和7.2Gy伽馬射線照射組的微核率明顯高于空白對照組(P均0.01);照射前給予S-異戊烯基-L-半胱氨酸可明顯降低1.0Gy與7.2Gy伽馬射線所致的微核率,分別由(39.5000±3.3141)‰降至(28.1667±4.1345)‰(P=0.033)和(76.5000±4.6242)‰降至(22.8333±3.6553)‰(P=0.000)。1.0Gy伽馬射線照射組和7.2Gy伽馬射線照射組的骨髓嗜多染紅細(xì)胞(PCE)與正染紅細(xì)胞(NCE)的比值(PCE/NCE)明顯低于空白對照組(P均0.05);照前給予S-異戊烯基-L-半胱氨酸可明顯升高相應(yīng)劑量組的PCE/NCE值(P均0.05)。結(jié)論 S-異戊烯基-L-半胱氨酸具有良好的DNA輻射損傷防護(hù)作用。
[Abstract]:Objective to evaluate the protective effect of S-isoamyl-L-cysteine on radiation-induced DNA damage in animal models of acute radiation injury. Methods 40 ICR mice were randomly divided into 1. 0 Gy 緯-ray irradiation group and 7. 2 Gy 緯-ray irradiation combined with Sisopentenyl-L- L- group in a control group with 1.0 Gy 緯-ray irradiation and 7. 2 Gy 緯-ray irradiation combined with Sisopentenyl-L- Cysteine administration group, 5 groups, Comet assay of peripheral blood lymphocytes and micronucleus assay of bone marrow polychromatic erythrocytes were used to detect DNA double strand breaks in mice of different treatment groups after exposure to 1.0 Gy and 7.2 Gy gamma rays in each group. Results DNA content, tail length, tail moment and Olive tail moment of peripheral blood lymphocytes in mice irradiated with 7.2 Gy gamma rays were significantly higher than those in control group (P 0.01). Tail length, The micronucleus rate of the tail moment and Olive tail moment was significantly lower than that of the control group (P 0.05). The micronucleus rate of the 1.0 Gy gamma ray group and the 7.2 Gy gamma ray group was significantly higher than that of the blank control group (P 0.01), and the ratio of Sisoamyl L-cysteine before irradiation was significantly higher than that of the control group (P 0.01).
