硫化氫對(duì)創(chuàng)傷應(yīng)激引起大鼠急性肝損傷的作用

【摘要】:目的探討硫化氫(H_2S)在擠壓大鼠后肢致急性肝損傷過程中的作用。方法將大鼠隨機(jī)分為對(duì)照組、擠壓組、H_2S供體硫氫化鈉(Na HS)+擠壓組、H_2S生成抑制劑炔丙基甘氨酸(propargylglycine,PAG)+擠壓組。用標(biāo)準(zhǔn)重物擠壓大鼠后肢建立急性肝損傷模型,分別于擠壓結(jié)束后30、120 min處死大鼠。比色法檢測(cè)血清天門冬氨酸氨基轉(zhuǎn)移酶(AST)、丙氨酸氨基轉(zhuǎn)移酶(ALT)活性,化學(xué)法檢測(cè)大鼠血漿H_2S、肝組織丙二醛(malondialdehyde,MDA)、蛋白質(zhì)羰基、谷胱甘肽(glutathion,GSH)含量和H_2S生成酶胱硫醚γ-裂解酶(cystathionineγ-lyase,CSE)活性,RT-PCR半定量檢測(cè)肝組織CSE m RNA的表達(dá)。結(jié)果擠壓大鼠后肢可引起血清AST、ALT活性增高,肝組織MDA、蛋白質(zhì)羰基含量增加及GSH含量降低,血漿H_2S含量減少,肝組織CSE活性降低、CSE m RNA表達(dá)下降。預(yù)先給予Na HS可顯著減輕、而PAG明顯加劇擠壓后肢所致上述肝損傷改變。結(jié)論 H_2S生成減少參與介導(dǎo)創(chuàng)傷應(yīng)激引起的大鼠急性肝損傷。
[Abstract]:Objective to investigate the role of hydrogen sulfide (H _ 2S) in the process of acute liver injury induced by extrusion of hind limbs in rats. Methods the rats were randomly divided into two groups: the extruded group with H2S donor sodium sulfohydride (NaHS) extruding group and the extruded group with propargyl glycine PAG (propargyl glycine) as an inhibitor of H2S production. The model of acute liver injury was established by pressing the hind limbs of rats with standard weight. The rats were killed at 30120 min after extrusion. The activities of aspartate aminotransferase (AST) and alanine aminotransferase (alt) were detected by colorimetric method. The plasma H2S, malondialdehyde (MDA) and protein carbonyl group were detected by chemical method in rat plasma. The content of glutathione (GSH) and the activity of cystathionine 緯 -lyase (cystathionine 緯 -lyase) were used to detect the expression of CSE mRNA in liver tissue. Results the results showed that the activity of ASTO alt in serum was increased, the content of protein carbonyl and GSH in liver tissue was increased, the content of H2S in plasma was decreased, the activity of CSE in liver was decreased and the expression of CSE mRNA in liver was decreased. Pretreatment with NaHS significantly alleviated the liver injury caused by compression of the hind limbs, while PAG significantly aggravated the changes. Conclusion the reduction of H2S production is involved in acute liver injury induced by traumatic stress in rats.
