【摘要】:目的:研究長(zhǎng)期運(yùn)動(dòng)性疲勞大鼠心肌膠原纖維形態(tài)結(jié)構(gòu)重塑及大蒜素的干預(yù)作用。方法:分別采用胃酶酸解法、苦味酸—天狼星紅染色法、逆轉(zhuǎn)錄聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)法、放射免疫技術(shù)、雙抗體夾心法、免疫組織化學(xué)法測(cè)定大鼠心肌羥脯氨酸(HYP)的含量(并計(jì)算膠原濃度(CC))、心肌膠原容積分?jǐn)?shù)(CVF)、小動(dòng)脈周?chē)z原容積分?jǐn)?shù)(PVCA)、心肌血管緊張素轉(zhuǎn)換酶(ACE)mRNA表達(dá)、心肌局部血管緊張素Ⅱ(AngⅡ)含量、血清Ⅰ型前膠原羧基端肽(PICP)、Ⅲ型前膠原氨基端肽(PIHNP)、心肌AT1受體(AT1R)及Ⅰ型和Ⅲ型膠原蛋白的表達(dá)水平。結(jié)果:疲勞組大鼠心肌羥脯氨酸(HYP)、膠原濃度(CC)、心肌膠原容積分?jǐn)?shù)(CVF)、小動(dòng)脈周?chē)z原容積分?jǐn)?shù)(PVCA)、心肌局部血管緊張素Ⅱ(AngⅡ)含量、血清Ⅰ型前膠原羧基端肽(PICP)、心肌血管緊張素轉(zhuǎn)換酶(ACE)mRNA表達(dá)、心肌AT1受體(AT1R)及Ⅰ型膠原蛋白表達(dá)量顯著升高,PIHNP及Ⅲ型膠原蛋白表達(dá)量顯著降低;干預(yù)組大鼠較疲勞組大鼠心肌羥脯氨酸(HYP)含量、膠原濃度(CC)、心肌膠原容積分?jǐn)?shù)(CVF)、小動(dòng)脈周?chē)z原容積分?jǐn)?shù)(PVCA)、心肌局部血管緊張素Ⅱ(AngⅡ)含量、血清Ⅰ型前膠原羧基端肽(PICP)、肌血管緊張素轉(zhuǎn)換酶(ACE)mRNA表達(dá)、心肌AT1受體(AT1R)及Ⅰ型膠原蛋白表達(dá)顯著降低,PIHNP及Ⅲ型膠原蛋白表達(dá)量顯著升高。結(jié)論:大蒜素干預(yù)能使長(zhǎng)期運(yùn)動(dòng)性疲勞大鼠心肌纖維化早期良性逆轉(zhuǎn),可能與ACE mRNA、AT1R表達(dá)水平的改變有關(guān)。
[Abstract]:Aim: to study the morphological and structural remodeling of myocardial collagen fibers and the intervention of allicin in long-term exercise-induced fatigue rats. Methods: gastric enzymatic acidolysis, picric acid-Sirius red staining, reverse transcriptase chain reaction (RT PCR), radioimmunoassay and double antibody sandwich method were used, respectively. The content of hydroxyproline (HYP) in myocardium of rats was determined by immunohistochemical method.
