【摘要】：為觀察4周跑臺(tái)運(yùn)動(dòng)對(duì)慢性不可預(yù)知性應(yīng)激(CUS)致抑郁大鼠行為學(xué)、血漿促腎上腺皮質(zhì)激素(ACTH)及皮質(zhì)醇(CORT)含量、海馬CA1、CA3區(qū)NGF表達(dá)的影響,將30只SD大鼠隨機(jī)分為對(duì)照組、應(yīng)激模型組及應(yīng)激運(yùn)動(dòng)組,大鼠建立慢性不可預(yù)見性應(yīng)激抑郁模型,同時(shí)應(yīng)激運(yùn)動(dòng)組大鼠進(jìn)行4周跑臺(tái)運(yùn)動(dòng)。運(yùn)動(dòng)及應(yīng)激結(jié)束后通過(guò)曠場(chǎng)實(shí)驗(yàn)、懸尾實(shí)驗(yàn)方法評(píng)定大鼠行為學(xué)指標(biāo)變化情況,采用八臂迷宮實(shí)驗(yàn)測(cè)試大鼠的學(xué)習(xí)記憶能力,使用放射免疫法測(cè)試大鼠血漿ACTH及CORT含量,采用免疫組織化學(xué)結(jié)合圖像半定量方法對(duì)海馬CA1、CA3區(qū)NGF神經(jīng)元的數(shù)量及面積進(jìn)行測(cè)量和分析。結(jié)果顯示:1)與應(yīng)激模型組相比,應(yīng)激運(yùn)動(dòng)組大鼠穿越格數(shù)、直立次數(shù)及修飾次數(shù)均顯著增加(P0.01,P0.05,P0.05),中央格停留時(shí)間顯著縮短(P0.05),懸尾不動(dòng)時(shí)間縮短(P0.05);2)與應(yīng)激模型組相比,應(yīng)激運(yùn)動(dòng)組大鼠完成八臂迷宮時(shí)間顯著縮短(P0.05),RME及TE均顯著減少(P0.05,P0.05);3)與應(yīng)激模型組相比,應(yīng)激運(yùn)動(dòng)組大鼠血漿ACTH及CORT含量顯著降低(P0.05,P0.01);4)與應(yīng)激模型組相比,應(yīng)激運(yùn)動(dòng)組大鼠海馬CA1區(qū)、CA3區(qū)NGF免疫陽(yáng)性細(xì)胞數(shù)量及面積均顯著增加(P0.05,P0.05,P0.05,P0.05)。得出結(jié)論:跑臺(tái)運(yùn)動(dòng)可改善抑郁模型大鼠的學(xué)習(xí)記憶能力,機(jī)理可能與長(zhǎng)期運(yùn)動(dòng)降低抑郁大鼠的血漿ACTH及CORT水平、拮抗HPA軸功能亢進(jìn)及增強(qiáng)海馬CA1區(qū)、CA3區(qū)NGF的表達(dá)有關(guān)。
[Abstract]:In order to observe the effects of treadmill exercise for 4 weeks on behavior, plasma adrenocorticotropic hormone (ACTH) and cortisol cortisone (Cort), and the expression of NGF in hippocampal CA1 / CA3 region, 30 SD rats were randomly divided into control group. The chronic unpredictable stress depression model was established in the stress model group and the stress exercise group, and the rats in the stress exercise group underwent treadmill exercise for 4 weeks. After exercise and stress, the changes of behavioral indexes in rats were evaluated by open field test and tail suspension test. The learning and memory abilities of rats were tested by eight arm maze test, and the contents of ACTH and CORT in plasma of rats were measured by radioimmunoassay. The number and area of NGF neurons in hippocampal CA1 / CA3 region were measured and analyzed by immunohistochemistry and image semi quantitative method. The results showed that compared with the stress model group, the rats in the stress exercise group were significantly increased in the number of traverses, the number of upright positions and the times of modification, the residence time of the central lattice was significantly shortened than that of the stress model group, and the suspended tail immobility time was shortened as compared with the stress model group. Compared with the stress model group, the plasma ACTH and CORT contents in the stress exercise group were significantly lower than those in the stress model group. In the stress exercise group, the number and area of NGF immunoreactive cells in the hippocampal CA1 region and CA3 region were significantly increased. Conclusion: treadmill exercise can improve the ability of learning and memory in depressive rats. The mechanism may be related to long-term exercise decreasing the levels of plasma ACTH and CORT, antagonizing the hyperfunction of HPA axis and enhancing the expression of NGF in CA3 region of CA1 region of hippocampus.
