模擬失重效應下龍血素B在大鼠體內的藥物動力學及排泄

【摘要】:比較正常重力與模擬失重效應下大鼠口服龍血素B的血漿藥物動力學及排泄差異.大鼠吊尾21d模擬失重效應,單次灌胃給予25mg/kg龍血素B.于系列時間點收集靜脈血,并分時間段收集尿液、糞便和膽汁,采用HPLC-MS方法測定各樣本中龍血素B含量,并求算藥物動力學參數(shù)和排泄量.模擬失重效應下龍血素B的血藥濃度-時間曲線下面積(AUC0-t)、血藥峰濃度(Cmax)、達峰時間(tmax)、表觀分布容積(Vd)、消除速率常數(shù)(Ke)以及生物半衰期(T1/2)與正常重力組相比均出現(xiàn)顯著變化,說明模擬失重效應明顯改變龍血素B在大鼠體內的藥物動力學過程.尿液、糞便和膽汁中龍血素B的排泄量也在增加,但與正常重力相比不明顯.
[Abstract]:The difference of plasma pharmacokinetics and excretion between rats with normal gravity and simulated weightlessness was compared. The weightlessness of rats was simulated by tail suspension for 21 days, and the rats were given 25mg/kg dragonin B by single gavage. Venous blood was collected at a series of time points, urine, feces and bile were collected in different time periods. The content of dragonin B in each sample was determined by HPLC-MS method, and the pharmacokinetic parameters and excretion quantity were calculated. Under simulated weightlessness, the area under the plasma concentration / time curve of dragonin B was significantly different from that of the normal gravity group, such as the area under the plasma concentration curve (AUC0-t), the peak concentration of C _ (max1), the peak time to the peak, the apparent distribution volume of VdN, the elimination of the rate constant (Keg) and the biological half-life (T _ (1 / 2)), as compared with the normal gravity group. The results showed that simulated weightlessness significantly changed the pharmacokinetic process of dragonin B in rats. Excretion of dragonin B in urine, feces and bile is also increasing, but not as significant as normal gravity.
