跳躍運(yùn)動(dòng)對(duì)大鼠脛骨骨調(diào)因子BMP2、cAMP的影響及電針干預(yù)作用

【摘要】:目的:本研究通過(guò)對(duì)生長(zhǎng)期大鼠進(jìn)行跳躍訓(xùn)練,采用電針作為直接干預(yù)手段,探討跳躍運(yùn)動(dòng)及電針對(duì)生長(zhǎng)期大鼠骨代謝的影響。從而為青少年運(yùn)動(dòng)員骨的正常生長(zhǎng)發(fā)育及骨損傷的預(yù)防與治療上提供一定的理論參考。 方法:40只一月齡Wistar大鼠,隨機(jī)分為空白對(duì)照組、跳躍運(yùn)動(dòng)組、電針預(yù)防組、電針治療組、自然恢復(fù)組五組,每組8只。實(shí)驗(yàn)采用電刺激跳躍訓(xùn)練,針刺干預(yù)選用腎俞、足三里、陽(yáng)陵泉三穴位。除空白組均進(jìn)行6周電刺激跳躍訓(xùn)練。電針預(yù)防組每次訓(xùn)練結(jié)束半小時(shí)后施以電針干預(yù)。電針治療組6周跳躍運(yùn)動(dòng)結(jié)束后休息一天再施以電針干預(yù),1次/天,6天/療程,中間休息一天,治療3個(gè)療程。光鏡觀察脛骨組織切片,雙抗體夾心ABC-ELISA法測(cè)試脛骨組織BMP-2和cAMP表達(dá)。 結(jié)果: (1)隨著時(shí)間的增加,大鼠體重呈增加趨勢(shì),但是空白組與預(yù)防組體重在6周實(shí)驗(yàn)過(guò)程中差異無(wú)統(tǒng)計(jì)學(xué)意義;空白組與跳躍運(yùn)動(dòng)組大鼠體重在第2周時(shí),前者明顯高于后者,且差異具有統(tǒng)計(jì)學(xué)意義(P0.01);預(yù)防組大鼠體重從第3周開始高于跳躍運(yùn)動(dòng)組大鼠體重,但從第4周開始預(yù)防組大鼠體重明顯高于跳躍運(yùn)動(dòng)組,且差異有統(tǒng)計(jì)學(xué)意義。 (2)6周跳躍運(yùn)動(dòng)訓(xùn)練后,HE結(jié)果顯示跳躍運(yùn)動(dòng)組較空白組板層骨內(nèi)膠原纖維排列明顯紊亂,骨皮質(zhì)密度較稀疏,骨細(xì)胞密集成團(tuán)狀,皮質(zhì)骨內(nèi)可見不規(guī)則,呈波紋狀的鈣化線。電針預(yù)防組較跳躍運(yùn)動(dòng)組大鼠脛骨板層骨內(nèi)膠原纖維排列稍有紊亂,外骨膜處無(wú)可見骨溶解破壞區(qū)和肉芽組織增生,骨干鮮見局灶性粘液變性區(qū),骨皮質(zhì)密度接近正常,骨細(xì)胞分布較均勻。電針治療組大鼠脛骨板層骨內(nèi)膠原纖維排列規(guī)則有序,皮質(zhì)骨內(nèi)骨細(xì)胞分布均勻,骨陷窩數(shù)量少,,哈佛氏管無(wú)擴(kuò)張、淤血,骨小梁排列規(guī)整有序,骨細(xì)胞分散均勻。 (3)跳躍運(yùn)動(dòng)組大鼠脛骨組織cAMP、BMP-2的表達(dá)明顯高于空白對(duì)照組與預(yù)防組,且具有顯著性差異(P0.01,P0.01); (4)治療組大鼠脛骨組織cAMP、BMP-2的表達(dá)明顯低于自然恢復(fù)組,且具有顯著性差異(P0.05,P0.01)。 結(jié)論: (1)根據(jù)HE染色結(jié)果提示6周的電刺激跳躍運(yùn)動(dòng)訓(xùn)練使生長(zhǎng)期大鼠脛骨出現(xiàn)了骨損傷,為進(jìn)一步研究跳躍運(yùn)動(dòng)對(duì)骨代謝的影響及骨損傷的研究提供了較好的動(dòng)物模型。 (2)一方面,跳躍運(yùn)動(dòng)可以促進(jìn)cAMP和BMP-2的表達(dá);另一方面,當(dāng)骨損傷發(fā)生時(shí)也能促進(jìn)cAMP和BMP-2的表達(dá)。提示跳躍運(yùn)動(dòng)引起的骨代謝變化與cAMP和BMP-2對(duì)骨組織的影響有關(guān)。 (3)電針干預(yù)具有雙向調(diào)節(jié)作用。一方面,電針干預(yù)可以降低跳躍運(yùn)動(dòng)對(duì)生長(zhǎng)期骨的不良影響,對(duì)骨損傷發(fā)生具有一定的預(yù)防作用;另一方面,當(dāng)骨損傷時(shí),電針又能對(duì)骨損傷進(jìn)行治療,且具有一定的療效。
[Abstract]:Aim: to investigate the effect of jumping exercise and electroacupuncture on bone metabolism in growing rats by using electroacupuncture as a direct intervention.It provides a theoretical reference for the normal growth and development of bone and the prevention and treatment of bone injury.Methods 40 one-month-old Wistar rats were randomly divided into five groups: blank control group, jumping exercise group, electroacupuncture prevention group, electroacupuncture treatment group and natural recovery group with 8 rats in each group.The experiment used electric stimulation jump training, acupuncture intervention selected Shenshu, Zusanli, Yanglingquan three acupoints.Except blank group, 6 weeks electric stimulation jumping training was performed.Electroacupuncture intervention was performed half an hour after each training in the electroacupuncture prevention group.In the electroacupuncture treatment group, one day of rest after 6-week jumping exercise was given, and then electroacupuncture intervention was given once a day / day for 6 days / course, and then one day for 3 courses of treatment.The expression of BMP-2 and cAMP in tibia tissue was measured by double antibody sandwich ABC-ELISA method.Results:1) with the increase of time, the body weight of the rats increased, but there was no significant difference between the blank group and the prevention group in the 6-week experiment, the weight of the blank group and the jumping group was significantly higher than that of the latter group at the second week.The body weight of the prevention group was higher than that of the jumping group from the third week, but the weight of the prevention group was significantly higher than that of the jumping exercise group from the 4th week, and the difference was statistically significant.After 6 weeks of jumping exercise training, the HE results showed that the collagenous fibers in the lamellar bone of the jumping exercise group were obviously disordered, the density of the bone cortex was sparse, the bone cells were dense and clustered, and irregular cortical calcification lines were observed in the cortex.Compared with the jumping exercise group, the collagenous fibers in the tibial lamina in the electroacupuncture prevention group were slightly disordered, no osteolysis and granulation tissue proliferation were found in the outer periosteum, focal mucinous degeneration was rarely seen in the diaphysis, and the density of the bone cortex was close to normal.The distribution of bone cells was more uniform.(3) the expression of cAMPP BMP-2 in tibial tissue in jumping exercise group was significantly higher than that in control group and prevention group, and the difference was significant (P 0.01).(4) the expression of cAMPP BMP-2 in the tibia of the treatment group was significantly lower than that in the natural recovery group, and there was significant difference between the two groups.Conclusion:1) according to the results of HE staining, 6 weeks of electric stimulation training made the tibia appear bone injury in growing rats, which provided a good animal model for further study on the effect of jumping exercise on bone metabolism and bone injury.2) on the one hand, jumping can promote the expression of cAMP and BMP-2, on the other hand, it can also promote the expression of cAMP and BMP-2 when bone injury occurs.The results suggest that the changes of bone metabolism induced by jumping exercise are related to the effects of cAMP and BMP-2 on bone tissue.3) electroacupuncture intervention has two-way regulating effect.On the one hand, electroacupuncture intervention can reduce the adverse effects of jumping exercise on the growth of bone, and has a certain preventive effect on bone injury; on the other hand, when bone injury, electroacupuncture can also treat bone injury, and has a certain effect.
