離心跑臺運動對小鼠骨骼肌炎性因子、肌再生因子及血管再生因子表達的影響

【摘要】:目的:觀察小鼠離心運動后骨骼肌炎性因子、肌再生因子以及血管再生因子的表達變化,探討其在離心運動骨骼肌損傷修復中可能發(fā)揮的作用。方法:32只雄性C57BL/6小鼠隨機分為對照組(C組,n=8)和離心運動組(D組,n=24)。根據取材時間,離心運動組又分為3個小組(運動后1d,3d和7d組)。離心運動后不同時間點取雙側腓腸肌。HE染色觀察骨骼肌形態(tài)學變化,比色法檢測血漿肌酸激酶(CK)和乳酸脫氫酶(LDH)活性,熒光定量PCR檢測炎性因子、肌再生因子及血管再生因子基因表達的變化。結果:小鼠下坡跑后骨骼肌纖維結構無明顯破壞,無顯著炎性細胞浸潤。與對照相比,血漿CK和LDH活性在運動后各時間點均無顯著變化(P0.05)。從基因層面看,炎性細胞標志物(MPO—嗜中性粒細胞標志物;F4/80—巨噬細胞標志物)在下坡跑后顯著上升。部分炎癥因子(如腫瘤壞死因子-α、白介素-1β、白介素-6)在下坡跑后顯著上調。而肌再生調節(jié)因子中,只有機械生長因子和白血病抑制因子在運動后出現顯著上調。而肌再生關鍵細胞——肌衛(wèi)星細胞的特異性標記分子(分子標志—Pax7,增殖標記—Myo D,分化標記—myogenin)在離心運動后未出現顯著變化(P0.05)。此外,其他與肌肉和血管再生密切相關的再生因子(如:胰島素樣生長因子1、肝細胞生長因子、尿激酶型纖溶酶原激活物、環(huán)氧合酶2、低氧誘導因子-1α、血管內皮生長因子、血管生成素1等)在離心運動后均未見顯著變化(P0.05)。結論:從形態(tài)學表現、血清指標和基因表達綜合來看,離心跑臺運動并非小鼠骨骼肌損傷建模的理想方式。
[Abstract]:Aim: to observe the expression of inflammatory factor, muscle regeneration factor and vascular regeneration factor in skeletal muscle after eccentric exercise in mice, and to explore the possible role of these factors in the repair of skeletal muscle injury after eccentric exercise.Methods 32 male C57BL/6 mice were randomly divided into control group (C group) and centrifugal exercise group (group D).According to the sampling time, the centrifuge exercise group was divided into 3 groups (1 d after exercise on 3 d and 7 d after exercise).The morphological changes of skeletal muscle were observed by HE staining at different time points after centrifugation. The activities of creatine kinase (CK) and lactate dehydrogenase (LDH) in plasma were detected by colorimetry, and inflammatory factors were detected by fluorescence quantitative PCR.Changes of gene expression of muscle regeneration factor and vascular regeneration factor.Results: there was no obvious destruction of skeletal muscle fiber structure and no significant inflammatory cell infiltration after running downhill in mice.Compared with the control, the activities of CK and LDH in plasma did not change significantly at each time point after exercise.At the gene level, the inflammatory cell marker, MPO- neutrophil marker, F4 / 80-macrophage marker, increased significantly after downhill running.Some inflammatory factors (such as TNF-偽, IL-1 尾, IL-6) were significantly up-regulated after downhill running.Among the regulating factors of muscle regeneration, only mechanical growth factor and leukemia inhibitory factor were significantly up-regulated after exercise.However, the specific labeling molecules (molecular marker -Pax7, proliferative marker -Myo D, differentiation marker -myogenin) of muscle regeneration key cell-mysatellite cells did not show significant changes after centrifugation.In addition, other regeneration factors closely related to muscle and vascular regeneration (such as insulin-like growth factor 1, hepatocyte growth factor, urokinase-type plasminogen activator, cyclooxygenase-2, hypoxia inducible factor -1 偽, vascular endothelial growth factor),Angiopoietin-1, et al.) there was no significant change in P0.05 after centrifugal exercise.Conclusion: centrifugal treadmill exercise is not an ideal way to model skeletal muscle injury in mice from morphological manifestation, serum index and gene expression.
