離心跑臺(tái)運(yùn)動(dòng)對(duì)小鼠骨骼肌炎性因子、肌再生因子及血管再生因子表達(dá)的影響

【摘要】:目的:觀察小鼠離心運(yùn)動(dòng)后骨骼肌炎性因子、肌再生因子以及血管再生因子的表達(dá)變化,探討其在離心運(yùn)動(dòng)骨骼肌損傷修復(fù)中可能發(fā)揮的作用。方法:32只雄性C57BL/6小鼠隨機(jī)分為對(duì)照組(C組,n=8)和離心運(yùn)動(dòng)組(D組,n=24)。根據(jù)取材時(shí)間,離心運(yùn)動(dòng)組又分為3個(gè)小組(運(yùn)動(dòng)后1d,3d和7d組)。離心運(yùn)動(dòng)后不同時(shí)間點(diǎn)取雙側(cè)腓腸肌。HE染色觀察骨骼肌形態(tài)學(xué)變化,比色法檢測(cè)血漿肌酸激酶(CK)和乳酸脫氫酶(LDH)活性,熒光定量PCR檢測(cè)炎性因子、肌再生因子及血管再生因子基因表達(dá)的變化。結(jié)果:小鼠下坡跑后骨骼肌纖維結(jié)構(gòu)無(wú)明顯破壞,無(wú)顯著炎性細(xì)胞浸潤(rùn)。與對(duì)照相比,血漿CK和LDH活性在運(yùn)動(dòng)后各時(shí)間點(diǎn)均無(wú)顯著變化(P0.05)。從基因?qū)用婵?炎性細(xì)胞標(biāo)志物(MPO—嗜中性粒細(xì)胞標(biāo)志物;F4/80—巨噬細(xì)胞標(biāo)志物)在下坡跑后顯著上升。部分炎癥因子(如腫瘤壞死因子-α、白介素-1β、白介素-6)在下坡跑后顯著上調(diào)。而肌再生調(diào)節(jié)因子中,只有機(jī)械生長(zhǎng)因子和白血病抑制因子在運(yùn)動(dòng)后出現(xiàn)顯著上調(diào)。而肌再生關(guān)鍵細(xì)胞——肌衛(wèi)星細(xì)胞的特異性標(biāo)記分子(分子標(biāo)志—Pax7,增殖標(biāo)記—Myo D,分化標(biāo)記—myogenin)在離心運(yùn)動(dòng)后未出現(xiàn)顯著變化(P0.05)。此外,其他與肌肉和血管再生密切相關(guān)的再生因子(如:胰島素樣生長(zhǎng)因子1、肝細(xì)胞生長(zhǎng)因子、尿激酶型纖溶酶原激活物、環(huán)氧合酶2、低氧誘導(dǎo)因子-1α、血管內(nèi)皮生長(zhǎng)因子、血管生成素1等)在離心運(yùn)動(dòng)后均未見(jiàn)顯著變化(P0.05)。結(jié)論:從形態(tài)學(xué)表現(xiàn)、血清指標(biāo)和基因表達(dá)綜合來(lái)看,離心跑臺(tái)運(yùn)動(dòng)并非小鼠骨骼肌損傷建模的理想方式。
[Abstract]:Aim: to observe the expression of inflammatory factor, muscle regeneration factor and vascular regeneration factor in skeletal muscle after eccentric exercise in mice, and to explore the possible role of these factors in the repair of skeletal muscle injury after eccentric exercise.Methods 32 male C57BL/6 mice were randomly divided into control group (C group) and centrifugal exercise group (group D).According to the sampling time, the centrifuge exercise group was divided into 3 groups (1 d after exercise on 3 d and 7 d after exercise).The morphological changes of skeletal muscle were observed by HE staining at different time points after centrifugation. The activities of creatine kinase (CK) and lactate dehydrogenase (LDH) in plasma were detected by colorimetry, and inflammatory factors were detected by fluorescence quantitative PCR.Changes of gene expression of muscle regeneration factor and vascular regeneration factor.Results: there was no obvious destruction of skeletal muscle fiber structure and no significant inflammatory cell infiltration after running downhill in mice.Compared with the control, the activities of CK and LDH in plasma did not change significantly at each time point after exercise.At the gene level, the inflammatory cell marker, MPO- neutrophil marker, F4 / 80-macrophage marker, increased significantly after downhill running.Some inflammatory factors (such as TNF-偽, IL-1 尾, IL-6) were significantly up-regulated after downhill running.Among the regulating factors of muscle regeneration, only mechanical growth factor and leukemia inhibitory factor were significantly up-regulated after exercise.However, the specific labeling molecules (molecular marker -Pax7, proliferative marker -Myo D, differentiation marker -myogenin) of muscle regeneration key cell-mysatellite cells did not show significant changes after centrifugation.In addition, other regeneration factors closely related to muscle and vascular regeneration (such as insulin-like growth factor 1, hepatocyte growth factor, urokinase-type plasminogen activator, cyclooxygenase-2, hypoxia inducible factor -1 偽, vascular endothelial growth factor),Angiopoietin-1, et al.) there was no significant change in P0.05 after centrifugal exercise.Conclusion: centrifugal treadmill exercise is not an ideal way to model skeletal muscle injury in mice from morphological manifestation, serum index and gene expression.
