耐力運動及環(huán)孢霉素A對大鼠骨骼肌纖維組成的影響

【摘要】:研究目的:通過耐力運動及注射環(huán)孢霉素A(CsA)干預(yù)鈣調(diào)神經(jīng)磷酸酶(CaN)的信號通路,探討對大鼠骨骼肌纖維組成的影響。 研究方法:以雄性SD大鼠為研究對象,分組進行適應(yīng)性訓(xùn)練,將SD大鼠隨機分為四組:對照組(C組)、CsA組(S組)、運動CsA組(ST組)、單純運動組(T組),每組10只。四組SD大鼠在第1~2周進行兩周的適應(yīng)性訓(xùn)練,第3~第15周進行耐力訓(xùn)練, S組、ST組在第13至15周進行皮下注射CsA15mg/kg/day(共3周)。在停止訓(xùn)練后的第二天取SD大鼠的比目魚肌和趾長伸肌,用SDS-PAGE電泳測肌球蛋白重鏈(MHC)組成。用南京建成的琥珀酸脫氫酶試劑盒測試SDH的活性和用定磷法測CaN的活性,并用Western blotting法測活化T細胞核因子(NFATc1)的蛋白含量。 研究結(jié)果: SDH: ST組與C組相比有非常顯著性升高(P<0.05);ST組與C組相比有非常顯著性升高(P<0.01)。 肌球蛋白濃度:大鼠比目魚肌(SOL)肌球蛋白濃度比較, S組與C組相比有顯著性下降(P<0.05);ST組與C組相比顯著性下降(P<0.05)。大鼠趾長伸(EDL)各組肌球蛋白的濃度無明顯差異。 CaN:大鼠比目魚。⊿OL)的CaN活性,S組與C組相比有顯著性下降(P<0.05);ST組與C組相比有非常顯著性下降(P<0.01);ST組與T組相比有顯著性下降(P<0.05)。大鼠趾長伸。‥DL)的CaN的活性, T組與C組相比有非常顯著性升高(P<0.01);ST組與S組相比有顯著性下降(P<0.05);ST組與T組相比有非常顯著性下降(P<0.01)。 MHC:大鼠比目魚肌MHCⅠ,ST組與C組相比具有顯著性升高,MHCⅡa顯著性下降(P<0.05)。大鼠趾長伸肌MHCⅡx, S組與C組相比有非常顯著性升高,MHCⅡd非常顯著性下降(P<0.01)。大鼠趾長伸肌MHCⅡx, ST組與S組相比具有非常顯著性下降MHCⅡd非常顯著性升高(P<0.01)。 NFATC1:大鼠比目魚肌NFATC1, T組與C組相比有顯著性下降(P<0.05);ST組與C組相比具有非常顯著性下降(P<0.01);ST組與S組相比有非常顯著性下降,,(P<0.01)。 研究結(jié)論: 1.注射CsA可導(dǎo)致大鼠飲食減少、體重下降,并伴有皮下組織囊腫和潰爛。 2.耐力運動增強了有氧氧化酶的活性,使大鼠骨骼肌SDH得到進一步的發(fā)展。CsA的干預(yù)并沒有影響SDH的活性變化。 3.耐力運動和注射CsA均可以引起大鼠慢肌比目魚肌的肌球蛋白降解。 4.注射CsA可以抑制CaN的活性,耐力運動可以引起快肌趾長伸肌的CaN活性提高,但對對慢肌比目魚肌的CaN活性沒有影響,說明不同肌纖維注射CsA干預(yù)CaN的程度不同。 5.耐力運動及注射CsA可以使慢肌比目魚肌的MHCⅠ增加、MHCⅡa減少,快肌趾長伸肌MHCⅡx減少、MHCⅡd增加。 6.耐力運動使慢肌比目魚肌中Ca2+濃度提高,引起CaN的活性提高,使大量NFATC1去磷化進入胞核內(nèi)。注射CsA抑制了CaN的表達使NFATC1沒有進入細胞核中。耐力運動及CsA共同作用使比目魚肌使胞漿中NFATC1大量入胞核。
[Abstract]:Objective: to investigate the effects of endurance exercise and cyclosporine (CsA) injection on the signal pathway of calmodulin phosphatase (Ca) in skeletal muscle of rats. Methods: male Sprague-Dawley rats were divided into groups for adaptive training. Sprague-Dawley rats were randomly divided into four groups: control group C, CSA group S group, exercise CsA group St group, exercise group T group with 10 rats in each group. Endurance training was conducted from week 3 to week 15, and group S was given subcutaneous injection of CSA 15 mg / kg / day at the 13th to 15th weeks. The soleus muscle and extensor digitorum longus muscle of SD rats were taken on the second day after stopping training. The composition of myosin heavy chain (MHC) was measured by SDS-PAGE electrophoresis. The activity of SDH was tested by the succinate dehydrogenase kit built in Nanjing, the activity of CaN was measured by the method of fixed phosphorus, and the protein content of activated T cell nuclear factor (NFATc1) was measured by Western blotting method. Results of the study:. SDH: there was a significant increase in SDH between St group and C group (P < 0.05). Myosin concentration: compared with group C, the concentration of myosin in group S was significantly lower than that in group C (P < 0.05) and that in group St was significantly lower than that in group C (P < 0.05). There was no significant difference in the concentration of myosin between group S and group C (P < 0.05), but there was no significant difference in the concentration of myosin between group S and group C (P < 0.05). The activity of CaN in group S was significantly lower than that in group C (P < 0.05). The activity of CaN in group St was significantly lower than that in group C (P < 0.01). The activity of CaN in extensor digitorum longus of rats was significantly decreased compared with group T, and the activity of CaN in group S was significantly lower than that in group C (P < 0.01). The activity of CaN in extensor digitorum longus in S group was significantly lower than that in group C (P < 0.05), and that in group T and T was significantly lower than that in group C (P < 0.01). There was a very significant increase in group C (P < 0.01) and a significant decrease in group S (P < 0.05) compared with group T (P < 0.01). MHC: compared with C group, MHC 鈪
